蒙药组方对大鼠乳腺上皮细胞增殖和分泌及细胞免疫功能的调节

摘  要:目的探讨蒙药组方治疗乳腺增生的机制。方法应用不同浓度蒙药组方刺激体外培养的乳腺上皮细胞,测定乳腺上皮细胞增殖及酪蛋白mRNA表达;建立大鼠乳腺增生模型,检测不同浓度蒙药组方对大鼠T淋巴细胞亚群的影响。结果蒙药组方可有效抑制乳腺上皮细胞的增殖,其中0.105mg/L组在有效抑制乳腺上皮细胞增殖的基础上,对酪蛋白mRNA表达无抑制作用;0.105g/kg蒙药组方CD4+/CD8+均值高于正常对照组和模型对照组,可以促进机体的细胞免疫功能,能扭转造模过程对机体免疫功能的抑制作用。结论蒙药组方可有效抑制大鼠乳腺上皮细胞增殖并能促进大鼠细胞免疫功能。Objective To study the mechanism of Compound Mongolia Medicine(CMM)on treating mammary gland hyperplasia.Methods Applying different concentration of CMM to stimulate mammary epithelial cells in vitro,the mammary cell proliferation and casein mRNA expression were tested;administrating a series dose of CMM to rat model of mammary gland hyperplasia and detecting the changes of CD4^+and CD8^+T lymphocyte to evaluate the function of cell-mediated immunity.Results 0.105mg/L CMM can effectively inhibit the proliferation of breast epithelial cell without inhibiting casein mRNA expression;and 0.105g/kg CMM can promote the cell-mediated immunity;CD4^+/CD8^+of 0.105g/kg CMM group is higher than that of normal control and mammary gland hyperplasia model.Conclusion CMM can effectively inhibit rat mammary epithelial cell proliferation and promote rat cell-mediated immunity.
