摘 要:目的:通过艾烟冷凝物对大鼠肺泡巨噬细胞NR8383的吞噬功能以及细胞活性氧的影响以探讨其对修复机体损伤和防卫免疫等方面是否具有一定调节的作用。方法:体外培养NR8383细胞,加入不同浓度的艾烟冷凝物,染毒24h后,应用流式细胞仪和DCFH-DA试剂盒来分别检测NR8383的细胞吞噬率及ROS。结果:在一定浓度范围内,艾烟冷凝物可以增强NR8383细胞的吞噬功能,各实验组(除浓度为0.004mg/ml的实验组外)与空白对照组的差异均具有统计学意义(P<0.005)。同时,在一定浓度范围内,它还可以降低NR8383细胞内的活性氧,分别对NR8383细胞用艾烟冷凝物染毒24h,该细胞的活性氧随着艾烟冷凝物的浓度升高而呈降低趋势,各实验组与空白对照组均具有统计学差异(P<0.001)。结论:在一定浓度范围内,艾烟冷凝物可以增强大鼠肺泡巨噬细胞NR8383的吞噬功能,亦可降低该细胞的活性氧,这可能成为艾烟能够修复机体损伤和提高自身免疫力的重要机制之一。Objective: To discuss that if the Moxa smoke had any regulatory role on the body's immune defense and repairing body injury by exploring the effects of the phagocytosis and reactive oxygen species on the rat's alveolar macrophages( NR8383) of the Moxa smoke condensate. Methods: The NR8383 cells were cultured in vitro. We put Moxa smoke condensate with different concentrations into a culture medium where the NR8383 cells were cultured,after the infected time of 24 hours,we observed the effects on the phagocytosis and ROS of the rat's alveolar macrophages by using the flow cytometry and the DCFH-DA kits. Results: Moxa smoke condensate can enhance the phagocytic function of the NR8383 cells. Compared with the controlled group,the experimental group except the 0. 004 mg/ml group were different statistically( P〈0. 005). With a certain concentration range,the Moxa smoke condensate can lower the reactive oxygen species of the NR8383 cells. After the different concentrations of Moxa smoke condensate expose the NR8383 cells for 24 hours,the ROS showed a decreasing trend with increasing the concentrations of the Moxa smoke condensate. Compared with the controlled group,the experimental group except the 0. 004 mg/ml group were different statistically( P〈0. 001). Conclusion: In a certain concentration range,Moxa smoke condensate can enhance the phagocytosis and reduce the ROS of the NR8383 cells with a certain concentration-dependent. This effect may become an important mechanism for increasing the body's immune ability,and repairing the damage by using moxibustion.
