寻常型银屑病血热证患者IL-6R/STAT3通路及其下游基因的表达

摘　　要：目的:探讨IL-6R/STAT3通路及其下游基因在寻常型银屑病血热证中的表达及意义。方法:采用RTPCR方法检测寻常型银屑病血热证患者及正常对照组外周血白细胞IL-6R、JAK2、STAT3、CD80、CCL5的表达。结果:上述检测指标相对表达量在疾病组中分别为0.333±0.079、0.206±0.040、0.296±0.048、0.219±0.053、0.588±0.114,与正常对照组表达量0.205±0.060、0.192±0.038、0.194±0.057、0.003±0.001、0.372±0.107比较,其中IL-6R、STAT3、CD80、CCL5表达水平明显高于正常对照组,差异有统计学意义(P<0.05,P<0.01),而JAK2表达无明显变化,差异无统计学意义(P>0.05)。结论:IL-6R/STAT3通路及其下游CD80、CCL5均可能参与了寻常型银屑病血热证的发病过程;IL-6R可能不是通过JAK2而是通过JAK家族中的其他途径或旁路来影响STAT3的表达。Objective：To study the expression and significance of the IL- 6R/STAT3 signaling pathway and downstream genes in blood heat syndrome patients with psoriasis vulgaris. Methods ： Set up the healthy controls and detect the expressions of IL - 6R, JAK2,STAT3 ,CD80 and CCL5 in the leukocyte of the blood heat syndrome patients with psoriasis vulgaris by the RT - PCR method. Results：The above detective indexes in disease group were 0. 333±0. 079,0. 206 ±0.040,0. 296 ±0. 048,0. 219 ±0. 053 and 0.588 ± 0.114. Compared with those in the control group （0. 205 ± 0.060,0. 192 ± 0. 038,0. 194 ± 0. 057,0. 003 ± 0. 001,0. 372 ± 0. 107）, the expression levels of IL - 6R, STAT3, CD80 and CCL5 were much higher with significant difference （ P 〈 0. 05, P 〈 0. 01 ）. There were no obvious changes in the expression of JAK2 or significant difference （P 〉 0. 05 ）. Conclusion：IL - 6R/STAT3 signaling pathway and CD80 as well as CCL5 may participate in the disease process of the blood heat syndrome patients with psoriasis vulgaris. IL - 6R has made an effect on STAT3 and it possibly achieves in different ways of other members of JAK or other routes but not JAK2.
