白芨环阿屯烷型三萜对血管生成和肿瘤细胞增殖的抑制作用

摘  要:研究白芨环阿屯烷型三萜对血管生成和肿瘤细胞增殖的抑制作用.通过大鼠动脉环血管生成模型观察白芨三萜对血管生成的抑制作用,通过MTT法检测白芨三萜对HeLa细胞增殖的抑制率,通过流式细胞术分析白芨三萜对细胞凋亡和细胞周期的影响.50μg/mL和100μg/mL的白芨三萜对大鼠动脉环微血管生成具有显著抑制作用.白芨三萜处理HeLa细胞后,其IC_(50)为61.33±1.78μmol/mL.白芨三萜阻止细胞于G1期,且具有诱导凋亡和坏死的双重作用.白芨三萜可抑制血管生成和肿瘤细胞增殖.This paper is to study the inhibitory effects of cycloartane triterpene from Bletilla striata on angiogenesis and tu- mor cell proliferation. The inhibitive effects of the triterpene on angiogenesis were observed by the rat aortic rings model. The inhibitive rates of the ttiterpene on HeLa cell proliferation were detected by MTT method. The influence of the triter- pene on cell apoptosis and cell cycle were analyzed by flow cytometry. 50 μg/mL and 100 μg/mL of the triterpene showed remarkable inhibition effects on microvessel formation of rat aortic tings. The IC50 of the triterpene on HeLa cells propaga- tion was 61.33 ~ 1.78 μmol/mL. The triterpene could block cell mitosis in G1 phase, and have the double roles of apopto- sis and necrosis by flow eytometry analysis. The triterpene from Bletilla striata had the inhibitive effects on angiogenesis and tumor cell proliferation.
