摘  要:目的研究雷公藤红素对非小细胞肺癌细胞株H1975的增殖抑制作用及相关机制。方法以不同浓度雷公藤红素(0.1、0.25、0.5、1.0、2.0μmol/L)分别作用于H1975细胞12 h或24 h,CCK-8法检测细胞增殖活性;小室侵袭法检测细胞的侵袭能力;Western blot法检测雷公藤红素对H1975细胞EGFR信号通路蛋白及其下游基因表达水平的调控作用;裸鼠成瘤体内验证雷公藤红素对H1975细胞增殖的抑制作用。结果雷公藤红素对H1975细胞增殖有较明显的抑制作用,且这种抑制作用呈浓度和时间依赖性;雷公藤红素能够显著抑制H1975细胞的侵袭能力;雷公藤红素对EGFR通路蛋白及其下游基因表达水平均有不同程度的抑制作用。结论雷公藤红素通过同时抑制EGFR及其下游信号通路蛋白的表达抑制H1975细胞的增殖与侵袭能力。Objective To study the inhibitory effect of celastrol on non-small cell lung cancer cell line H1975 and its mechanism. Methods H1975 cells were treated with different concentrations of celastrol(0.1, 0.25, 0.5, 1.0, 2.0 μmol/L)for 12 h or 24 h respectively. The proliferation activity of H1975 cells was detected by CCK-8. The invasiveness of the cells was detected by chamber invasion assay. Western blotting was used to detect the regulatory effect of celastrol on the expression of EGFR signaling pathway protein and downstream genes in H1975 cells. The inhibitory effect of celastrol on the proliferation of H1975 cells was verified inside nude mice with tumor. Results Celastrol showed significant inhibitory effect on the proliferation of H1975 cells, and such inhibitory effect was concentration-and time-dependent;celastrol was able to significantly inhibit the invasiveness of H1975 cells; celastrol showed different degrees of inhibitory effects on the expression of EGFR pathway proteins and downstream genes. Conclusion Celastrol inhibits the proliferation and invasion of H1975 cells by inhibiting the expression of EGFR and downstream signaling pathway protein.
