中药复方糖肾安对实验性2型糖尿病大鼠肾脏ROS水平及Nrf2、HO-1表达的影响

摘　　要：目的探讨中药复方糖肾安对实验性2型糖尿病大鼠肾脏ROS水平、Nrf2、HO-1表达的影响。方法70只健康SD大鼠随机分为正常组(n=10)和造模组(n=60)。造模组大鼠高糖高脂饮食4周后一次性腹腔注射STZ(35 mg/kg)建立2型糖尿病大鼠模型,将成模的大鼠随机分为模型组、糖肾安低、中、高剂量组、西药组。药物治疗8周后,用流式细胞仪检测ROS水平,用定量RT-PCR、Western blot检测Nrf2、HO-1表达情况。结果各治疗组ROS水平均高于正常组(P<0.01),糖肾安组ROS水平均明显低于模型组(P<0.01)。各治疗组Nrf2、HO-1表达均低于正常组(P<0.01),糖肾安中、高剂量组的表达较高,高于模型组(P<0.01)。结论中药复方糖肾安可能通过激活抗氧化应激Nrf2/ARE通路,诱导糖尿病大鼠肾脏Nrf2、HO-1的表达,降低ROS水平,改善肾脏炎症反应,从而起到肾脏保护作用。Objective To discuss the effect of Tang Shenan decoction on ROS levels and Nrf2,HO-1 expression in experimental type 2 diabetic rats kidney. Methods The 70 healthy SD rats were randomly divided into normal group（ n = 10） and manufacturing model group（ n = 60）. The manufacturing model group by high-lipids and high-sucrose diets for 4th week and then intraperitoneal injection STZ（ 35 mg·kg） to establish the diabetic rats model,then they were randomly divided into model group,low,medium and high Tang Shenan decoction group,western medicine group,each group according to the corresponding drug lavage. Eight weeks later,the Flow cytometry analysis ROS levels,by quantitative RT-PCR,Western blot test Nrf2,HO-1 expression. Results Each treatment group ROS levels were higher than normal group（ P 0. 01）,Tang Shenan decoction group ROS levels were lower than the model group（ P 0. 01）. Nrf2 and HO-1 expression in each treatment group were lower than normal group（ P 0. 01）,the expression of medium and high Tang Shenan decoction group is higher,higher than that of model group（ P 0. 01）.Conclusion Tang Shenan decoction may through activation of Nrf2 / ARE antioxidant stress,induced diabetic rats kidney expression of Nrf2,HO-1,reduce the ROS levels,improve kidney inflammation and protection kidney.
