疏肝饮对内脏高敏感性大鼠结肠中TRPA1及PAR2表达的影响

摘　　要：目的观察疏肝饮对内脏高敏感性大鼠结肠组织中TRPA1和PAR2表达的影响。方法 36只Wistar雄性大鼠随机分为正常组、模型组、疏肝饮大剂量组、疏肝饮中剂量组、疏肝饮小剂量组、西药组,采用慢性避水应激法造模10天,造模第4天起,各给药组大鼠于造模前半小时分别给予药物灌胃,连续7天,第10天造模结束后,处死大鼠,取出结肠组织,采用realtime-PCR及免疫组化法分别检测TRPA1和PAR2含量。结果与正常组相比,模型组结肠中TRPA1和PAR2的含量显著增加(P<0.001或P<0.01);与模型组比较,疏肝饮组大、中剂量组和得舒特组大鼠结肠中TRPA1和PAR2含量均明显减少,差异有统计学意义(P<0.001或P<0.01)。结论疏肝饮可能通过调节结肠中TRPA1和PAR2的表达,从而降低内脏高敏感性。Objective To investigate the effects of Shugan - decoction on TRPA1 and PAR2 in the colon of rats with visceral hy- persensitivity. Methods Thirty - six male wistar rats were randomly divided into six groups as control group, model group, low dose Shugan - decoction group, middle dose Shugan - decoction group, High dose Shngan - decoction group and dicetel group. Rats were exposed to 1 - h water avoidance stress （WAS） daily for 10 consecutive days. Shugan - decoction groups and dicetel group were given intragastric administration half hour before WAS from the 4th day to the 10th day. The animals were sacrificed and colonic tissues collected at the end of the 10 - day period, immunohistochemistry and real - time polymerase chain reaction were applied to investigate the content of TRPA1 and PAR2 in the colon. Results Compared with the normal group, TRPA1 and PAR2 in the colon of model group were significantly increased ; Compared with the model group, TRPA1 and PAR2 in the colon of middle dose Shugan - decoction group, High dose Shugan - decoction group and dicetel group were significantly reduced. Con- clusion The mechanism of Shugan - decoction reduce visceral hypersensitivity may involve mediating the expressions of TRPA1 and PAR2 in colon tissues.
