摘  要:目的建立同时测定新疆维药芹菜根提取物中3种黄酮类成分含量的HPLC方法。方法 poroshell EC-C_(18)色谱柱(4.6 mm×150 mm,2.7μm),流动相为乙腈-0.1%磷酸(20∶80),流速0.6 ml·min^(-1),检测波长340 nm,柱温28℃,进样量5μl。结果芹菜苷、3'-甲氧基芹菜苷、芹菜素-7-O-β-D-吡喃葡萄糖苷的质量分别在0.046~0.274μg(r=0.9999),0.025~0.153μg(r=0.9999),0.010~0.062μg(r=0.9998)内与峰面积线性关系良好,加样回收率均值分别为97.81%,97.02%,99.85%,RSD为1.81%,1.50%,1.61%。结论表明所建立的方法简便、快速、准确、重现性好,可用于新疆维药芹菜根提取物中黄酮类成分的含量测定。Objective A HPLC method was established to simultaneously determine the contents of apiin, 3"- methoxy apiin and apigenin - 7 - O - β - D - glueopyranoside in the extract of Apium graveolens L. root in Xinjiang. Methods The HPLC methodwas performed on an Agilent poroshell EC - C18 column (4.6 mm × 150 mm, 2.7 μm) eluted with the mobile phase of water con- sisting 0.1% phosphoric acid and acetonitrile in gradient mode. The UV detection wavelength was set at 340 nm. The flow rate was 0.6 ml · min^- 1 and the column temperature was controlled at 28 ℃. Results All the standard compounds showed a good lin- earity in the range of 0.046 - 0.274 μg( r = 0. 9999 ) for apiin, 0. 025 - 0. 153 μg( r = 0. 9999 ) for 3"- methoxy apiin, 0. 010 -0. 062 μg( r = 0. 9998 ) for apigenin - 7 - O - β - D - glucopyranoside. The average recoveries of these three ilavonoids were 97.81%, 97.02% and 99.85% with RSD 1.81%, 1.50% and 1.61%, respectively. Conclusion In conclusion,the method is simple, rapid and accurate, which was suitable for the quantitative analysis of three kinds of flavonoids in the extract of Apium graveolens L. root.
