摘 要:目的通过离体与在体实验分别观察天麻素对长春新碱抑制乳腺癌细胞MCF-7生长和抗Walker-256乳腺癌荷瘤大鼠肿瘤生长作用的影响。方法体外培养乳腺癌细胞株MCF-7,采用MTT法检测不同剂量长春新碱对MCF-7细胞存活率的影响,计算长春新碱抑制乳腺癌细胞MCF-7生长的IC_(50),再测定单独给予不同浓度天麻素及其分别于IC_(50)长春新碱联合孵育24 h后MCF-7细胞生长的抑制率。通过SD大鼠在体皮下接种Walker-256乳腺癌细胞建立乳腺癌模型;实验分为A组:对照组;B组:乳腺癌肿瘤模型组;C组:乳腺癌肿瘤模型+长春新碱组;D、E组:乳腺癌肿瘤模型+长春新碱+60 mg/kg或120 mg/kg天麻素组。观察各组大鼠肿瘤生长情况,称瘤重计算抑瘤率,采用HE染色观察肿瘤切片组织形态学变化。结果长春新碱抑制乳腺癌细胞MCF-7生长的IC_(50)为28.34μg/mL;与空白对照组比较,不同浓度天麻素对MCF-7细胞的生存率无明显的改变(P>0.05);与单用长春新碱组比较,不同浓度天麻素与长春新碱联合给药对MCF-7细胞的生存率无明显的变化(P>0.05)。在在体实验中,与A组比较,B组大鼠肿瘤生长明显,瘤重达到32.76 7.65 g;与B组比较,C组肿瘤明显缩小,瘤重减轻到(19.30±4.24)g(P<0.05),抑瘤率41.09%;与C组比较,D、E组肿块进一步缩小,瘤重分别为(15.28±3.42)g、(10.99±2.87)g(P<0.05),抑瘤率分别达53.35%和66.44%。瘤体组织HE染色观察结果显示,B组肿瘤细胞核质比增高,核深染,可见病理性核分裂;与B组比较,C组核质比例减小,核呈圆形淡染,可见细胞坏死区域;与B组比较,D组中细胞坏死区域明显增大,可见部分肿瘤细胞核消失,E组细胞坏死进一步加重,可见片状凝固性坏死组织。结论天麻素对长春新碱抑制乳腺癌细胞MCF-7生长的作用无明显影响,但在体实验中天麻素能显著增强长春新碱对Walker-256荷瘤大鼠的抗肿瘤疗效,且呈剂量依赖性。Objective To investigate the effects of Gastrodin on Vincristine - induced growth inhibition of MCF - 7 breast cancer cells and rat Walker -256 tumor in vitro and in vivo. Methods In vitro experiments, MTT method was used to detect the half maximal inhibitory concentration of Vincristine - induced growth inhibition of human breast cancer MCF - 7 cells ; The effect of different concentration of Gastrodin on the growth inhibition rate of Vincristine in human breast cancer cell were evaluated. In vivo experiments, to establish rat tumor model through injecting subcutaneously Walker - 256 breast cancer cells into the rat armnit.rats were randomly divided into groups: A: normal control group; B: model control group; C: Vincristine group; D, E groups: Vincristine + 60mg / kg or 120mg / kg Gastrodin group. Twenty three days later, rats were euthanized, and tumors were re- moved and weighted, then calculated the tumor inhibition rate, HE staining was applied to observe the morphologic changes of tumor. Results The IC50 of Vincristine was 28. 341xg,/mL, the different concentration of Gastrodin did not decrease the growth of human breast cancer cell MCF - 7 ( vs. Normal control, P 〉 0.05 ), without affecting the rate of Vincristine in human breast cancer cell MCF - 7 ( vs. Vincristine group, P 〉 0.05 ) , the tumor weights in B group were 32.76 7.65 g, the tumor of C group was significant shrinked, the weights of tumor were only 19.3 4.24 g ( vs. B group, P 〈 0.05 ), the rate of tumor inhibition was 41.09% ; the tumor weights of D and E group were 15.28 3.42g, 10.99 2.87g, and tumor inhibition rate were 53.35% , 66. 44% , respectively. HE staining showed increased the ratio of tumor cell nucleus to mass, nuclear stained and visible pathological mitotic in B Group; when compared with B group, the proportion of nuclear of tumor in C Group reduced, nuclear circular stai- ning slighted ; However, D and E groups visible nuclei of tumor cells disappeared, D group of a certain cell necrosis, whereas,
