健脾复方对体外培养的大肠癌细胞SW620生长影响的最佳浓度筛选

摘　　要：目的观察健脾复方对体外培养大肠癌细胞SW620生长的影响。方法制备BALB/C裸小鼠含药血清,将大肠癌细胞SW620按空白对照组、药物空白组、不同浓度中药组(2.5%、5%、7.5%、10%、15%)在96孔板中进行培养,处理72h后,观察细胞形态,并采用CCK8法测定各组OD值,确定最佳浓度。结果 15%浓度中药组的OD值与空白对照组相似,差异无统计学意义(P>0.05),且低于药物空白组,差异有统计学意义(P<0.05),其余各浓度中药组OD值与药物空白组比较,差异无统计学意义(P>0.05)。结论健脾复方抑制体外培养的大肠癌细胞SW620生长的最佳浓度应为15%。Objective To observe the effect of Jianpi Compound Recipe on the growth of colorectal cancer cells SW620 in vitro culture. Methods Medicine containing serum of BALB/C nude mice was prepared, the colorectal cancer cells SW620 were cul- tured in 96 well plates dividing into blank control group, drug - free group, and different concentrations of the Chinese herbs group （ 2.5% ,5% ,7.5%, 10% , 15% ）. After 72 hours treated, observing the cell morphology, confirming the optimum concentration by determining the OD values with CCK8 method. Results The OD value of the 15% concentration Chinese herbs group is similar to that of the blank control group, the difference was no statistically significant （ P 〉 0.05 ）, and lower than that of the drug - free group,the difference was statistically significant（P 〈 0. 05 ） ,in the meanwhile, the differences of OD values between the rest con- centration Chinese herbs groups and drug - free group showed no statistically significant （ P 〉 0.05 ）. Conclusion The optimum concentration of Jianpi Compound Recipe inhibiting the growth of colorectal cancer cells SW620 in vitro culture should be 15%.
