摘 要:目的 高效液相色谱法测定云厚朴生品及3种炮制品中4种活性成分的含量,考察炮制对云厚朴主要化学成分的影响,为云厚朴资源的合理开发利用提供依据。方法 高效液相色谱法测定含量,色谱条件为:戴安U3000系列高效液相色谱仪(配备PDA检测器);Agilent SB-C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相:采用甲醇-0.1%冰醋酸梯度洗脱;流速:1.0 mL/min;检测波长:紫丁香苷为265 nm,丁香酚为280 nm,厚朴酚及和厚朴酚为294 nm;柱温:25℃,提取方法为甲醇超声提取。结果 所建立的4种活性成分含量测定方法平均加样回收率范围97.21%~98.36%,RSD在1.16%~1.43%范围内。4种活性成分的含量由高到低顺序为和厚朴酚、厚朴酚、紫丁香苷和丁香酚,但姜炙品中紫丁香苷含量低于丁香酚含量,生品及炮制品的厚朴酚及和厚朴酚的总含量满足2015年版《中国药典》的要求。结论 所建立的云厚朴生品及3种炮制品中4种活性成分含量测定方法准确、可靠。炮制方法的不同,对云厚朴药材活性成分的含量造成不同程度的影响。Objective The study aimed to determine the quantity of four active compounds in four samples of Magnolia rostrata W. W. Smith by HPLC - PDA, and to investigate the impact of processing on chemical composition. Methods An HPLC - PDA method was established to determine the contents of four constituents, syringin, eugenol, magnolol, honokiol by Dionex U3000 HPLC with PDA detector. The HPLC separation was carried out on an Agilent SB - Cls column (4.6 mm ×250 mm, 5 μm) with a mixture solution of methanol - acetic acid (0.1% ) as the mobile phase in a gradient elution at a flow rate of 1.0 mL/min. The wavelength was set at 265 nm for syringing, 280 nm for eugenol, 294 nm for magnolol and honokiol. The column temperature was set at 25 ℃. The active compounds were extracted by ultrasonic method in methanol. Results The average recovery of the four ac- tive compounds ranged from 97.21% to 98.36% with RSDs less than 2.00%. The contents of honokiol, magnolol, syringin, eu- genol in samples decreased in turn, but the content of syringin in the sample processed by ginger juice was less than of eguenol. Contents of honokiol and magnolol in four samples met the requirement in Chinese Pharmacopoeia (2015 edition ). Conclusion The HPLC method established is accurate and reliable. The processing may change the contents of the four active compounds in Magnolia rostrata W. W. Smith.
