益智仁盐炙有效部位中圆柚酮大鼠在体肠吸收

摘　　要：目的:考察盐炙对益智仁有效部位圆柚酮吸收的影响,研究益智仁盐炙"生熟有别"的内涵。方法:以酚红为标示物,圆柚酮为指标性成分,采用大鼠单向肠灌流模型,运用HPLC法测定肠灌流液中药物的浓度,比较炮制前后药物吸收参数。结果:益智仁有效部位圆柚酮在小肠各肠段吸收较好;不同质量浓度和p H的吸收速率参数(Ka)与有效渗透系数(Peff)值无显著性差异;益智仁盐炙前后圆柚酮Ka和Peff有显著性差异。结论:益智仁有效部位圆柚酮小肠吸收较好,不受质量浓度的影响,提示其吸收机制主要为被动扩散吸收;炮制后圆柚酮吸收增大,可能是盐炙后能增强止尿作用的机理之一。Objective： To study the effects of nootkatone of effective parts from salt processing of Alpiniae oxyphyllae Fructus（ AOF,益智仁） by rat intestinal absorption behavior of effective parts from in rats,complete scientific connotation of AOF processed. Method： With phenol red as the indicator,in situ rat single pass perfusion model was used and effective parts concentration intestinal segment was determined by HPLC with Nootkatone as the marker. To investigate the effects of concentration,p H,intestinal section on the absorption of the effective part,then compare the absorption parameters of AOF in raw and processed. Result： Nootkatone of effective part from AOF was well absorbed at all segments of the intestine. The absorption rate constant（ Ka） and effective permeability values（ Peff） were of different concentrations and p H were no significant difference（ P〈0. 05）,but the result of AOF processed was significantly affected（ P〈0. 05）. Conclusion： Nootkatone of effective part of AOF was well absorbed at all segments of intestine in rat,suggesting passive diffusion mechanism might be involved in the absorption of the effective part. The values of Ka and Peff were increased after processing,and it is one of mechanism to enhance the effect of reducing urine after salt processing.
