摘  要:目的探讨椎间盘退变大鼠模型中缺氧诱导因子1α(HIF-1α)因子水平的变化及其临床意义。方法选取椎间盘退变大鼠模型40只作为观察组,另选择健康大鼠模型40只作为对照组。选择两组大鼠的腰椎间盘组织经冲洗、超声粉碎、离心等一系列过程取得标本,采用酶联免疫吸附法检测两组腰椎间盘组织中的HIF-1α因子水平。结果观察组中每只大鼠的退变腰椎间盘组织中均有HIF-1α的表达,但表达含量存在明显的不同。HIF-1α在观察组中的表达含量为(25.12±4.02)pg/g,HIF-1α在对照组中的表达含量为(7.84±2.19)pg/g,HIF-α在观察组的表达含量明显高于对照组,差异具有统计学意义(P<0.05)。结论 HIF-1α在腰椎间盘退变大鼠模型的腰椎间盘组织中的表达水平较高,推测其可能参与了人类的椎间盘退变过程,具有一定的临床意义。Objective To explore HIF-1α factor changes of intervertebral disc degeneration in the rat model.Methods 40 rats with intervertebral disc degeneration were selected as observation group,another 40 healthy rats model were selected as control group.To choose two groups of rats model of lumbar organization by flushing,ultrasonic grinding,centrifugal and a series of process to obtain serum specimens and the change of HIF- 1 alpha factor in lumbar organization were observed by the method of enzyme- linked immunosorbent.Results The expression of HIF alpha in lumbar disc degeneration organization were found in each rat of observation group,but there were distinct different in expression levels.The expression levels of HIF- 1α in the observation group were 25.12 ±4.02 pg/g and in the control group 7.84 ± 2.19 pg/g.There was statistically significant difference(P〈 0.05).Conclusion The expression of HIF- la in lumbar disc degeneration organization is high.It may be involved in the human intervertebral disc degeneration process,and has a certain clinical significance.
