摘 要:目的研究鬼箭锦鸡儿乙醇提取物(CE)的抗血栓作用。方法按照体重将雄性Sprague-Dawley(SD)大鼠随机分为阴性对照组(1%羧甲基纤维素钠)、阳性对照组(100 mg·kg^(-1)阿司匹林)和低、中、高3个剂量(CE,150,300,600mg·kg^(-1))实验组,每组8只。所有药物均用1%CMC-Na混悬,连续灌胃7 d,每天1次,给药体积为10μL·g^(-1)。末次给药2 h后,用氯化铁溶液诱导大鼠颈总动脉血栓模型。用全自动血液凝固分析仪测定凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)和凝血酶时间(TT);用酶标仪测定CE对大鼠血清一氧化氮(NO)和血浆P-选择素水平的影响。将Kunming(KM)小鼠按体重随机分为5组:阴性对照组和阳性对照组同上,低、中、高3个剂量(CE,200,400,800mg·kg^(-1))实验组,给药体积为20μL·g^(-1),其他用法同上。于末次给药2 h后,用毛细管法测定小鼠凝血时间。结果大鼠实验:与阴性对照组对血栓的抑制率为20.61%相比,低、中、高3个剂量实验组对血栓的抑制率分别为42.68%,54.07%,80.11%,差异有统计学意义(P<0.001)。与阴性对照组的PT为(10.74±0.46)s相比,中、高2个剂量实验组的PT分别为(12.60±0.87),(12.01±0.70)s,差异有统计学意义(P<0.01);与阴性对照组的APTT为(64.11±6.27)s比较,中、高2个剂量实验组的APTT分别为(70.34±2.86),(74.13±3.39)s,差异有统计学意义(P<0.05,P<0.01);与阴性对照组的TT为(26.89±2.22)s比较,高剂量实验组的TT为(33.57±1.50)s,差异有统计学意义(P<0.01)。与阴性对照组的血清NO水平为(5.17±0.28)μmol·L^(-1)比较,中、高2个剂量实验组的NO水平分别为(6.30±0.58),(6.55±1.62)μmol·L^(-1),差异有统计学意义(P<0.05);与阴性对照组的P-selectin水平为(2.08±0.50)μmol·L^(-1)比较,中、高2个剂量实验组的P-selectin水平分别为(1.61±0.38),(1.33±0.40)μmol·L^(-1),差异有统计学意义(P<0.05,P<0.01)。小鼠实验:与阴性对照组的小鼠凝血时间为(96.40±19.05)s比较,中�Objective To study the antithrombotic effects of Caragana jubata( Pall.) Poir. ethanol extract( CE) on carotid artery thrombosis in rats. Methods Male Sprague-Dawley( SD) rats were randomly divided into five groups: negative control group( 1% sodium carboxymethyl cellulose),positive control group( aspirin 100 mg · kg-1),experimental groups in low,medium and high doses( CE 150,300,600mg·kg-1,respectively),each group had eight rats. All drugs weresuspended in 1% CMC-Na and given orally once daily( 10 μL·g-1) for 7 days. Two hours after the last administration,carotid artery thrombosis models in rats were induced by ferric chloride. The prothrombintime( PT),activated partial thromboplastin time( APTT) and thrombin time( TT) were measured by Full Automatic Blood Coagulation Analyzer. Nitric oxide( NO) in rat serum and P-selectin in rat plasma were measured by Microplate Reader. Kunming( KM) mice were randomly divided into five groups: negative control group and positive control group( the same as in rat above),experimental groups in low,medium and high dose( CE 200,400,800 mg·kg-1,respectively). All drugs were given orally 20 μL·g-1. Other operations were the same as in rat experiments. Two hours after the last administration,clotting time experiment was conducted by capillary method. Results Rat experiments: Compared with the inhibition rate of 20. 61% in negative control group,CE has a significant inhibition rate of 42. 68%,54. 07%,80. 11% at the dose of 150,300,and 600 mg · kg-1respectively( P〈0. 001). The medium and high doses of CE could prolong PT to( 12. 60 ± 0. 87) s and( 12. 01 ± 0. 70) s obviously( P〈0. 01) compared with the negative control group with( 10. 74 ± 0. 46) s,and prolong APTT to( 70. 34 ± 2. 86) s and( 74. 13 ± 3. 39) s evidently( P〈0. 05,P〈0. 01) in contrast to the negative control group with( 64. 11 ± 6. 27) s.
