摘 要:探讨白藜芦醇通过调节甲状腺激素代谢通路中的关键基因SIRT1、PGC1α进而缓解机体氧化应激,改善甲状腺激素及其相关受体水平,调节能量代谢维持机体稳态。C57雄性小鼠随机分成3组,每周监测体质量,分别测定血脂、甲状腺激素以及氧化应激指标。与C组相比,HFD组小鼠体重、血脂显著升高(P<0.05);HFD组小鼠肝脏GSH/GSSG和T-AOC在第7周末显著降低,MDA显著上升(P<0.05);HFD组T4以及FT4水平与C组相比显著升高(P<0.05),T3以及FT3水平差异不显著;添加白藜芦醇能显著上调DIOI、TRβ、SIRT1、PGC1α的表达量。The research aimed to study the impact of high-fat diet on thyroid hormone receptor and deiodinase in mice, as well as the protective effect ofresveratrol. Four-week-old C57BL/6 male mice were fed normal diet, high-fat diet (HFD), or HFD supplemented with 0.1% resveratrol, respectively, metabolic experiments were conducted in the 6th week, the mice were sacrificed at the end of the 7th week and oxidative stress biomarker, blood lipid and thyroid hormone were then examined. The gene expressions of SIRT1, PGC1α, DIOI and TRβ were analyzed by RT-PCR. Compared with group C, body weight, blood lipid, MDA, T4 and FT4 levels of HFD mice were significantly increased (P 〈 0.05) whereas liver GSH/GSSG and T - AOC were remarkably reduced. Resveratrol obviously increased the expressions of DIOI, TR beta, SIRTI and PGC 1 alpha. Therefore, in a long term of high-fat feeding, the oxidative stress interfered with thyroid hormone but resveratrol could improve the metabolic abnormality by elevating the expressions of related genes SIRT1 ,PGC1α, DIOI and TRβ.
