摘  要:目的初步探讨SIRT1基因沉默对高脂喂养C57BL/6J幼鼠肝脏的影响。方法选取4周龄的雄性C57BL/6J小鼠32只,分为4组:高低脂饮食组及慢病毒注射组。在高脂饮食喂养6周后,尾静脉分别注射SIRT1shRNA慢病毒(LV-SIRT1 shRNA)和对照慢病毒(LV-control shRNA),继续高脂饮食1周。Western Blot检测肝脏中SIRT1表达情况,HE和油红O染色观察肝脏脂滴堆积情况。结果 Western Blot显示LV-SIRT1 shRNA慢病毒小鼠肝组织中SIRT1表达水平显著下降;肝组织HE和油红O染色显示,LV-SIRT1 shRNA组较LV-control shRNA组脂肪肝更明显。结论慢病毒介导SIRT1基因沉默可以加重高脂饮食诱导的非酒精性脂肪肝风险。Objective To investigate the effect of SIRT1 gene silencing on the liver tissue of high - fat :- induced C57BL/6J im- mature rats. Methods The male C57BL/6J mice (4 -old -week,n =32) were selected and divided into low -fat diet (LFD) ,high -fat diet (HFD) and lentivirus - injected groups. After all of them were HFD - fed for 6 weeks, the LV - SIRT1 shRNA or LV - control shRNA were injected into the tail vein of these I-IFD -fed mice separately,then the mice were HFD -fed for 1 week continously. Then the SIRT1 protein expression level of liver was detected by western blotting. And the accumulation of liver morphology and lipid droplets in liver were observed by H&E or Oil red O staining. Results Western blotting showed that SIRT1 expression level was significantly decreased in the LV - SIRT1 shRNA group. The H&E or Oil red O staining showed that comparing with the LV - control shRNA group, the level of lipid droplets synthesis and steatosis in the LV - SIRT1 shRNA group significantly decreased. Conclusions Lentivirus - mediated SIRTI gene silencing could aggravate the risk of non - alcoholic fatty liver disease (NAFLD) induced by HFD.
