摘 要:为获得纯合的GHR基因敲除香猪的种群,本研究以贵州从江香猪为母本,中国农业大学提供的GHR基因敲除猪(GHR(+/-))为父本杂交选育F1代,筛选出F1代阳性杂合子猪和阴性猪,再用F1代阳性杂合子猪进行扩繁得到F2代。测定F1代和F2代猪的生长指标,同时检测F1代和F2代阳性猪。实验结果表明F1代GHR基因敲除阳性猪(GHR+/+)和阴性猪(GHR+/-)生长指标差异均不显著(p>0.05)。F2代GHR基因敲除猪在3月龄时体长、体重存在显著差异(p<0.05),F2代4月龄阳性猪与阴性猪各项指标均差异显著(p<0.05)。测序结果显示F1阳性杂合子猪共11头,阴性猪15头;F2阳性纯合子猪共8头,阴性猪9头。In order to obtain the homozygous GHR gene knockout pig population, we selected hybridization breeding of F1 generation pig by Guizhou Congjiang pigs (GHR+/+) as the female parent and china agricultural university provide GHR gene knockout pig (GHR+/-) as the male parent, then we obtained F2 homozygous GHR gene knockout pig (GHR-/-) by crossbreeding of F1 generation pigs tested positive (GHR+/- male and GHR+/- female pigs). Afterwards, we detected the growth performance of the F 1 generation pigs and F2 generation pigs. The result showed that the growth performance of F1 generation pigs tested positive (GHR+/-) had lower than F1 generation pigs tested negative (GHR+/+), and no significant difference (p〉0.05). For hybrid F2 pigs, body length and body weight of GHR-/- pig had lower than GHR+/+ pigs in three-month-old, significant difference (p〈0.05). All the indexes of F2 generation GHR-/- pigs had lower than F2 generation GHR+/+ pigs in four-month-old, significant difference (p〈0.05). DNA sequencing result showed that GHR+/- and GHR+/+ of the hybrid F1 pigs have 11 and 15 respectively, GHR-/- and GHR+/+ of the hybrid F2 pigs have 8 and 9 respectively.
