摘  要:本研究以湘豆3号大豆种子为外植体探究消毒条件与消毒效果间关系,以胚尖为接种材料,建立了湘豆3号胚尖再生体系,以卡那霉素为筛选剂,获得抗生素选择最适浓度。研究表明:70%的酒精消毒1min后配合5%的NaClO消毒10min为湘豆3号大豆种子最佳的消毒方法;在MS+0.1mg/LTDZ的培养基中胚尖丛生芽诱导率最高,达到96.7%;不定芽在不添加生长素的1/2MS培养基中能正常生根,生根率达到76.67%;在MS培养基中添加浓度为75-100mg/L的卡那霉素为最适宜湘豆3号抗生素选择压力。Seeds of Xiangdou No.3 were used as explants to study the relationship between disinfection condition and sterilization effect. In this research we used the embryonic tips as inoculation materials to establish the regeneration system of Xiangdou No.3, Kanamycin as screening agent and choose the optimal concentration of antibiotics. The results showed that disinfecting with 70% alcohol for 1 minute with 5% NaClO sterilizing for 10 minutes was the best way of disinfection in Xiangdou No.3. The induction rate of embryonic tips of adventitious buds were the highest in the culture medium of MS with 0.1 mg/L TDZ, reaching to 96.7%. The rooting rate of adventitious buds reached 76.7% in 1/2 MS culture medium without adding any auxin. The selection pressure of kanamycin ranges from 75 to 100 mg/L which was added into MS culture medium, was the most appropriate for embryonic tips in Xiangdou No.3.
