黄柏不同炮制品对甲亢型肾阴虚模型大鼠甲状腺和肾上腺皮质功能的影响

摘　　要：目的:比较生黄柏、酒黄柏和盐黄柏对甲亢型肾阴虚模型大鼠甲状腺和肾上腺皮质功能的影响。方法:将88只大鼠随机分为空白组、模型组、黄酒组、盐水组、阳性组(六味地黄丸,0.2 g/kg,以生药计)以及生黄柏、酒黄柏、盐黄柏的高、低剂量组(剂量均为9.519、1.058 g/kg,以生药计),每组8只。除空白组外,其余各组大鼠均ig左旋甲状腺素钠复制肾阴虚模型;于造模同时各给药组大鼠ig相应药物,其余组大鼠ig等体积相应溶液,每天1次,连续3周。末次给药后测定大鼠体质量、24 h尿量、脏器(胸腺、脾)指数,并测定血清中游离三碘甲状腺原氨酸(FT3)、游离四碘甲状腺原氨酸(FT4)、抗利尿激素(AVP)、环磷酸腺苷(c AMP)、环磷酸鸟苷(c GMP)水平及尿液中17-羟皮质类甾醇(17-OHCS)水平,并计算c AMP/c GMP比值。结果:与空白组比较,模型组大鼠体质量、24 h尿量、脏器(胸腺、脾)指数均明显降低;血清中FT3、FT4、AVP、c AMP水平、c AMP/c GMP比值及尿液中17-OHCS水平明显升高,血清中c GMP水平明显降低,差异均有统计学意义(P<0.01)。除酒黄柏低剂量组大鼠体质量和酒黄柏高、低剂量组大鼠血清中c GMP水平外,阳性组及生、酒、盐黄柏的高、低剂量组大鼠上述指标较模型组均明显改善(P<0.05或P<0.01),改善程度大小依次为盐黄柏组>生黄柏组>酒黄柏组;且盐黄柏高剂量组大鼠血清中c AMP水平和c AMP/c GMP比值及盐黄柏高、低剂量组尿液中17-OHCS水平均明显低于同剂量生黄柏组(P<0.05)。结论:黄柏对甲亢型肾阴虚模型大鼠甲状腺和肾上腺皮质功能均有一定的改善作用,且盐制品作用增强,而酒制品作用减弱,提示黄柏给盐制后其滋阴效果有所增强。OBJECTIVE：To compare effects of Phellodendron chinense,stir-baked P. chinense with yellow rice wine and stirbaked P. chinense with salt solution on thyroid gland and adrenocortical function of kidney-Yin deficiency model rats with hyperthyroidism. METHODS：88 rats were randomly divided into blank group,model group,yellow rice wine group,salt solution group,positive group（Liuwei dihuang pills,0.2 g/kg,calculated by crude drug）,high- dose and low- dose groups of P. chinense,stirbaked P. chinense with yellow rice wine and stir- baked P. chinense with salt solution（9.519,1.058 g/kg,calculated by crude drug）. Except for blank group,other groups were given levothyroxine sodium to induce kidney-Yin deficiency model;during modeling,treatment groups were given relevant medicine intragastrically,and other groups were given constant volume of solution intragastrically,once a day,for consecutive 3 weeks. After last administration,body weight,24 h urine amount,viscera（thymus,spleen） indexes were determined;the serum levels of free triiodothyronine（FT3）,free thyroxine（FT4）,vasopressin（AVP）,c AMP and c GMP were determined as well as urine levels of 17-hydroxy steroid（17-OHCS）. c AMP/c GMP ratio was also calculated. RESULTS：Compared with blank group,body weight,24 h urine amount and viscera（thymus,spleen） indexes were decreased significantly in model group;serum levels of FT3,FT4,AVP and c AMP,c AMP/c GMP ratio and urine level of 17-OHCS were increased significantly,while serum level of c GMP was decreased significantly,with statistical significance（P〈0.01）. Ex-cept for the body weight in stir-baked P. chinense with yellow rice wine group and the serum level of c GMP in high-dose and low- dose of stir- baked P. chinense with yellow rice wine group,above indexes of positive group,high- dose and lowdose groups of P. chinense,stir-baked P. chinense with yellow rice wine and stir-baked P. chinense with salt solution were allimproved significantly（P〈0.05 or P〈0.01）.
