摘  要:目的:研究疏风解毒胶囊对急性支气管炎大鼠模型的保护作用,并探讨其对于炎性因子分泌的影响。方法:SPF级雄性SD大鼠60只,随机均分为对照(A)组、模型(B)组、疏风解毒胶囊治疗(C)组、地塞米松治疗(D)组。除A组外,余3组每天烟雾刺激2次,早晚各1次,共1周,制备成急性支气管炎大鼠模型。C组每天灌胃给药疏风解毒胶囊100mg/kg、D组每天腹腔注射地塞米松5mg/kg,于成模后第1、3、5天每组各处死5只,取其左主支气管组织作以下测试:HE染色病理学观察;气管组织IL-1β、IL-6免疫荧光标记后激光共聚焦显影。经颈静脉采血行Elisa检测IL-1β、TNF-α、Cxcl10及Ccl3表达水平。结果:B组大鼠呼吸道上皮炎性反应明显,其呼吸道上皮组织在成模后第1天时出现明显的渗出炎性反应,第3、5天时呼吸道上皮组织炎性反应达到高峰伴有明显的纤毛紊乱,部分组织坏死脱落;B组大鼠血清IL-1β、IL-6、TNF-α、Cxcl10及Ccl3水平较A组明显升高(P<0.05)。C组可明显缓解大鼠急性呼吸道上皮组织的损伤,其呼吸道上皮组织病理与B组比较,成模后第1、3、5天时呼吸道上皮组织的炎性渗出程度较轻。C组大鼠血清IL-1β、IL-6、TNF-α、Cxcl10及Ccl3水平较B组有明显下降(P<0.05);而C组与D组相比,两组大鼠血清IL-1β、IL-6、TNF-α、Cxcl10及Ccl3水平无明显差异。结论:疏风解毒胶囊可减轻急性支气管炎大鼠模型呼吸道上皮细胞炎性因子的分泌,缓解大鼠的急性呼吸道上皮炎性反应。Objective:To observe the protective effects of Shufeng Jiedu Capsule(SFJDC) on rat model of acute bronchitis and to explore its influence in the secretion of inflammatory factors in respiratory epithelium.Methods:Sixty SPF SD rats were randomly divided into 4 groups:control(A) group,model(B) group,Shufeng Jiedu Capsule(C) group,and Dexamethasone(D) group,with 15 rats in each group.Except for the A group,the others were subjected to smoke twice a day for a total of 1 week,in order to induce the rat model of acute bronchitis.The C group was given the administration of SFJDC by oral gavage at a dose of 100mg/kg,and the D group was given the administration of Dexamethasone by intraperitoneal injection at a dose of 5mg/kg.At the 1st day,3rd day and 5th day after the modeling,5 rats randomly chosen from each group were sacrificed to harvest their left bronchus tissues.HE staining was used to observe the pathological manifestation.The confocal laser scanning microscope technology was used to observe the fluorescence labeling of IL-1β and IL-6.Elisa was used to detect the levels of IL-1β,TNF-α,Cxc110 and Ccl3 in jugular vein blood.Results:The results of pathological manifestation showed that the inflammatory infiltration was obviously observed in the respiratory epithelial tissue in the B group when the rat model of acute bronchitis was successfully established for 1 day,where the inflammatory infiltration reached a peak,companied with remarkable inordinate cilia and tissue necrosis when the model was successfully established for 3 or 5 days.The levels of IL-1β,IL-6,TNF-α,Cxcl10 and Ccl3 in the serum of B group were significantly higher than those of the A group(P〈0.05).But,the administration of SFJDC significantly attenuated the acute damage in the respiratory epithelial tissue,and furthermore the C group showed a slighter inflammatory infiltration in respiratory epithelial tissue when compared with the B group at the 1 st day,3rd day and 5th day after the modeling.The levels of IL
