针刺百会、人中穴对急性脑缺血大鼠模型NF-κB/IκB-α的影响

摘　　要：目的:探讨针刺对急性脑缺血大鼠海马细胞核因子-κB(NF-κB)、核因子κB抑制因子-α(IκB-α)表达的影响。方法:3月龄SD雌性大鼠100只,随机均分为正常组、假手术组、穴位组、非穴位组、模型组。实验结束后取脑组织,TTC染色测定梗死面积,放免法检测缺血脑组织白介素-6(IL-6)和白介素-1β(IL-1β)含量,免疫组织化学法检测海马NF-κB、IκB-α的表达。结果:与正常组比较,模型组大鼠脑梗死范围明显,脑缺血组织中IL-6和IL-1β显著升高,海马CA3区NF-κB蛋白表达增加,IκB-α蛋白表达降低(P<0.01);与模型组相比,穴位组大鼠梗死区域变小,脑缺血组织中IL-6和IL-1β含量降低,海马CA3区NF-κB蛋白表达降低,IκB-α蛋白表达增加(P<0.05),但假手术组与模型组比较差异无统计学意义。结论:针刺可以显著增强NF-κB蛋白表达,抑制IκB-α的蛋白表达,从而减轻其诱导的各种炎性细胞因子的表达,从而发挥抗脑缺血损伤作用。Objective：Observation of acupuncture on acute cerebral ischemia rat in the hippocampus NF-κB,IκB-αexpression.Methods：One hundred female SD rats,aged three months,were randomly divided into five groups：normal group,sham-operation group,model group,sham acupuncture group,acupuncture group.We obtained brain tissues after treatment,and used TTC staining to obtain the cerebral infarction,adopted radioimmunoassa method to detect IL-6 and IL-1β content in ischemic tissue,took immunohistochemical detection on hippocampal NF-κB and IκB-α expression.Results：Compared to normal group,obvious infarction happend in model group,IL-6 and IL-1β in brain ischemic tissue became significantly higher,hippocampal NF-κB immunoreactive material MOD value was significantly higher,while IκB-α became lower（P〈0.01）.Compared to model group,brain infarction area was decreased in acupuncture group,IL-6 and IL-1β in brain ischemic tissue became significantly lower,hippocampal NF-κB immunoreactive material MOD value was significantly decreased while IκB-αwas increased（P0.05）.However,no significant difference between sham acupuncture group and model group.Conclusion：Acupuncture can significantly enhance NF-κB activity and inhibit protein IκB-α expression,thereby reduce the induction of various inflammatory cytokines and play a key role in anti-cerebral ischemic injury.
