摘 要:目的:利用核磁共振测定技术探讨维药溃结安对异常黏液质溃疡性结肠炎(UC)模型大鼠的干预效果,为UC的个体化治疗提供实验依据。方法:将Wistar大鼠随机分为UC溃结安干预(A)组和UC病症溃结安干预(B)组。其中,A组运用DNCB复合乙酸法建立UC模型大鼠,在此基础上用溃结安干预21d。B组以湿寒饲料(芫荽实和菠菜实),在温度(6±1)℃,相对湿度85%-95%的环境中进行喂养,建立异常黏液质证载体模型大鼠后运用DNCB复合乙酸法建立异常黏液质UC模型大鼠,在此基础上用溃结安干预21d。对两组模型大鼠进行鉴定,并收集血清,进行核磁共振氢谱(1H-NMR)测定,运用正交偏最小二乘判别(OPLS-DA)分析所采集的图谱,确认差异性代谢组分。结果:与A组比较,B组大鼠血清中乳酸、丙氨酸含量降低,β-羟丁酸、丙酮酸、乙酰乙酸含量升高(P<0.05)。结论:维药溃结安对异常黏液质UC大鼠的干预效果优于对UC大鼠的干预效果。Objective:To investigate the effects of Kuijie'an on rat model of ulcerative colitis(UC) with abnormal balgam carrier by nuclear magnetic resonance(NMR) technology,in order to provide experimental evidence for the individualized treatment of UC.Methods:Male Wistar rats were randomly divided into UC Kui Jie'an treatment group(group A) and abnormal balgam carrier UC Kui Jie'an treatment group(Group B).The rats in the Group A suffered the DNCB andacetic acid stress to stablish the rat model of UC.Then,the Kui Jie'an was used to treat the model for 21 days.The rats in the Group B were fed with wet and cold diet(seeds of spinach and Parsley) in a cold(6±1)℃ and humid(85%-95%) atmosphere to establish the abnormal balgam carrier model,which then suffered the DNCB andacetic acid stress.Then,the Kui Jie'an was used to treat the model for21 days.The serum samples were collected to conduct the ~1H-NMR analysis.Then,the data was analyzed by orthogonal partial squares-discriminant analysis(OPLS-DA) software to identify discrepant metabolic components.Results:Compared with Group A,the levels of lactic acid,alanine in the Group B were reduced,while the levels of β-hydroxybutyric acid,pyroracemic acid and acetacetic acid were increased(P〈0.05).Conclusion:The effects of Uyghur medicine Kui Jie'an on rat model of UC with abnormal balgam are superior to the effects on the rat model of UC.
