摘  要:目的研究空腹血糖受损的2型糖尿病前期人群和2型糖尿病新发患者尿液代谢指纹谱的变化模式,筛选出可能与2型糖尿病相关的潜在代谢标志物。方法利用气相色谱-质谱联用(Gas Chromatograph-Mass Spectrometer,GC-MS)技术对正常人群、空腹血糖受损的2型糖尿病前期人群和2型糖尿病新发患者尿液代谢指纹谱进行测定;运用多元统计方法对GC-MS数据进行分析,筛选出与2型糖尿病可能相关潜在代谢标志物。结果 2型糖尿病新发患者尿液代谢指纹谱的个体差异较正常人群和前期人群增大;正常人群、前期人群和新发患者尿液代谢指纹谱具有差异;初步筛选出三种可能与2型糖尿病相关的潜在代谢标志物D-氨基葡萄糖酸(Glucosoxime)、棕榈酸(Hexadecanoic acid)和β-假尿苷(β-pseudouridine)。结论本研究初步筛选出了可能与2型糖尿病相关的潜在尿液代谢标志物,证实了GC-MS技术联合偏最小二乘判别分析(Partial Least Squares-Discriminant Analysis,PLS-DA)多元统计分析应用于2型糖尿病早期发现和疾病状态监控的可行性。Objective To study the change patterns of metabolite fingerprints of urine in people with impaired fasting glucose and new patients with type 2 diabetes, and to select potential metabolic biomarkers of type 2 diabetes. Methods Gas chromatograph - mass spectrometer ( GC - MS) was used to analyze the metabolite fingerprints of urine. The data obtained from GC - MS was analyzed by multivariate statistical analysis, for selecting the potential metabolic biomarkers of type 2 diabetes. Results The individual differences of metabolite fingerprints of urine in new patients with type 2 diabetes increased compared to healthy people,people with impaired fasting glucose. There were differences in the metabolite fingerprints among the three groups. Three different potential metabolic biomarkers that may be relevant with type 2 diabetes were selected, including glucosamine,hexadecanoic acid, and β - pseudouridine. Conclusion In this study, we select several potential metabolic biomarkers that may be relevant with type 2 diabetes and confirm the feasibility of GC - MS coupled with PLS - DA applying in the early diagnosis of type 2 diabetes and monitoring of the disease progression.
