均匀试验法优选玄参总环烯醚萜苷提取工艺  

Single Factor-uniform Design Method Optimize the Extraction Technics of Scrophularia Total Iridoids

摘　　要：目的:研究玄参中以哈帕俄苷、桃叶珊瑚苷、哈帕苷为代表的总环烯醚萜苷最佳提取工艺。方法:采用HPLC测定含量,以玄参总环烯醚萜苷(STI)提取率为指标先通过单因素筛选对STI影响较大的因素,再进行均匀试验,确定最优工艺。结果:结合实际情况,STI最佳工艺为:乙醇浓度70%,提取温度85℃,溶剂量1∶14,提取次数2次,第1次8倍量,第2次6倍量,每次1 h,STI提取率为1.88%。结论:此工艺基本可行,可为工业大生产提供参考。Objective： To study the best extraction technics of Scrophularia total iridoids, typical with harpagoside, harpagide, ancubin. Methods： The rational extraction technic was estabilished by a combination of single-factor and uniform design experiment, using the yield of total iridoids as the evaluation marker, determined by HPLC. Results： The optimum process was obtained as follows： the crushed plant materials were extracted with 70% ethanol in 85 ℃ water bath temperature under reflux for twice, first with 8 times volumn of the plant materials for 1 hour, then with 6 times for another hour, the verification tests show that the yield of STI was 1.88% averagely under this condition. Conclusion： The technic was feasible, which is a reasonable reference to industrial production.
