摘  要:目的观察MMP-2/Notchl转染嗅鞘细胞(0EG)移植对急性大鼠脊髓损伤的作用。方法将构建的MMP-2/Notchl的质粒导入嗅鞘细胞,并移植入急性脊髓损伤大鼠体内,连续观察12周,与接受单纯OEG移植的脊髓损伤大鼠进行比较。结果移植转染MMP-2/Notchl能够增强嗅鞘细胞的迁移能力,移植到脊髓损伤大鼠体内与对照组比较,能减少组织的缺损、轴突的坍塌、神经元的坏死凋亡,抑制胶质细胞的过度增殖。结论MMP-2/Notchl转染的嗅鞘细胞相对于单独转染嗅鞘细胞能更有效的促进脊髓损伤功能的修复。Objective To investigate the therapeutic effect of olfactory ensheathing glia (OEG) transfected with MMP-2 / Notch1 gene on acute spinal cord injury. Methods The plasmid of MMP-a/Notchl were transfected into cultured OEGs and trans- planted into spinal cord injury in rats,observing for 12 weeks,which were compared with only OEG transplantation in rats. Results The transplanted OEGs transfected with MMP-2/Notchl gene not only reduce tissue defects, axon collapse, but can also inhibit neuronal apoptosis and necrosis and excessive proliferation of glial cells compared with control groups after spinal cord injury. Conclusion MMP-2/Notchl transfected OEGs can be more effectively promoting spinal cord Injury repair in rat.
