摘 要:目的探讨枸杞多糖(LBP)对糖尿病大鼠阴茎海绵体氧化应激损伤的保护作用。方法 SD大鼠腹腔注射链脲佐菌素(STZ)建立糖尿病大鼠模型,随机设立糖尿病组(DM组)、枸杞多糖灌胃组(LBP组),并设立正常对照组(NC组)。8周后通过APO试验检测勃起功能,硫代巴比妥酸法检测阴茎海绵体组织内丙二醛(MDA)含量,黄嘌呤氧化酶法检测阴茎海绵体组织内超氧化物歧化酶(SOD)含量,光镜下观察阴茎海绵体组织Masson染色病理切片中胶原纤维、平滑肌纤维的差异。结果与NC组比较,DM组大鼠勃起次数明显减少(P<0.05),阴茎海绵体组织内MDA含量显著增高(P<0.05),SOD含量明显减少(P<0.01);大鼠阴茎海绵体组织平滑肌部分断裂,数目减少,胶原纤维菲薄,结构破坏,排列紊乱。经灌胃8周后,LBP组与DM组比较:勃起次数增多(P<0.05),大鼠阴茎海绵体组织中MDA含量降低(P<0.05),SOD含量明显增加(P<0.01),光镜下观察阴茎海绵体组织结构改善。结论实验证实,LBP可以减轻糖尿病大鼠阴茎海绵体氧化应激损伤,修复部分组织结构,改善大鼠勃起功能。Objective To investigate the protective effects of Lycium barbarum polysaccharides (LBP) on oxidative stress in corpus cavemosum of diabetic rats. Methods The diabetic rat models were first induced by injection of streptozotocin (STZ), and then randomly divided into the diabetes group and the treated group with LBP. The normal rats were raised as normal control group. Eight weeks later, erectile function of rats was assessed by subcutaneous injection with apomorphine. The level of malondialdehyde (MDA) and the activites of superoxide dismutase (SOD) in cavernous tissue were detected. The Masson staining was used to show the alteration of fibre structure in rat penis. Results Compared with that of the normal control group, the level of MDA was increased remarkably in cavernous tissue of rats in the diabetes group, whereas the activities of SOD were decreased significantly. Meanwhile, a significant decrease of erection frequency was also found in the diabetic rats. Compared with that of diabetes group, the level of MDA was decreased significantly in cavernous tissue of rats in the treated group with LBP, whereas the activities of SOD were increased remarkably. Additionally, a remarkable reduction in smooth muscle fibers and collagen fibers in cavernous tissue of the diabetic rats, as well as disrupted continuity and integrity in smooth muscles were observed in Masson staining analysis. However, the structures of cavernous tissue were improved in the treated group with LBP. Conclusion This study suggested that Lycium barbarum polysaccharides could ameliorate injury of erectile function in DM rats by inhibiting oxidative stress in cavernous tissue.
