摘 要:[目的]本试验旨在研究亮氨酸对超早期断奶宫内发育迟缓(IUGR)仔猪内源消化酶活性及胰腺抗氧化能力的影响。[方法]从正常分娩的16头母猪中选择正常初生体质量(NBW)和IUGR新生仔猪各16头,所有仔猪于14日龄断奶。试验采用2×2因子设计,NBW仔猪和IUGR仔猪分别饲喂基础日粮(含1.45%亮氨酸)或亮氨酸日粮(含1.80%亮氨酸),即NC(NBW+基础日粮)、NL(NBW+亮氨酸日粮)、IC(IUGR+基础日粮)与IL(IUGR+亮氨酸日粮)共4组,每组8头猪,35日龄屠宰取样。[结果]IC组胰腺、十二指肠、空肠和回肠的淀粉酶活性显著低于其他组(P<0.05),IL组胰腺的淀粉酶活性显著低于NC组(P<0.05);IC组胰腺、十二指肠和回肠的脂肪酶活性显著低于其他组(P<0.05),IL组十二指肠的脂肪酶活性显著高于NC组(P<0.05);IC组胰腺的糜蛋白酶活性显著低于其他组(P<0.05),IC组十二指肠的糜蛋白酶活性显著低于IL组(P<0.05)。与NC组和IL组相比,IC组胰腺SOD、GPx活性和GSH含量显著降低(P<0.05),MDA含量显著升高(P<0.05)。与NC组相比,IC组胰腺SOD1 mRNA的表达水平显著降低(P<0.05),而GPx1、GPx7、CAT mRNA表达水平差异不显著(P>0.05);另外,IL组胰腺SOD1、CAT mRNA的表达水平显著高于IC组(P<0.05)。[结论]IUGR会导致早期断奶仔猪内源消化酶活性、胰腺中抗氧化酶活性和非酶抗氧化物质含量降低及抗氧化酶mRNA表达下调,引起机体消化吸收功能紊乱,而亮氨酸对胰腺的抗氧化功能和胃肠道的消化吸收功能有一定的改善作用。[ Objectives ] The purpose of this study is to investigate the effects of leucine on activities of endogenous digestive enzymes and pancreas antioxidant capacity in early weaning pigs suffering from intrauterine growth retardation (IUGR). [ Methods ] Sixteen normal body weight(NBW) piglets and sixteen IUGR piglets were selected according to birth weight. At weaning( 14 days of age), both NBW and IUGR pigs were fed a control diet( containing 1.45% leucine)or a leucine diet( containing 1.80% leucine). Thus, all pigs were randomly distributed into groups of 4 treatments : NC ( NBW + control diet), NL ( NBW + leucine diet), IC ( IUGR + control diet) and IL ( IUGR+leucine diet) for 21 d ( n = 8). [ Results ] The amylase activity of pancreas, duodenum, jejunum, and ileum in the IC group was significantly lower than in the other groups(P〈0.05) and the amylase activity of pancreas in the IL group was significantly lower than in the NC group(P〈0.05) ;the lipase activity of pancreas, duodenum, and ileum in the IC group was significantly lower than in the other groups(P〈0.05)and the lipase activity of duodenum in the IL group was significantly higher than in the NC group (P〈0.05) ; the chymotrypsin activity of pancreas in the IC group was significantly lower than in the other groups ( P〈0.05), and the chymotrypsin activity of duodenum in the IC group was significantly lower than in the IL group(P〈0.05). Compared with NC and IL groups, the activities of SOD and GPx, as well as the GSH content significantly decreased( P〈0.05), and the MDA content significantly increased (P〈0.05)in the pancreas of the IC group. Compared with NC group, the expression of SOD1 mRNA in the pancreas of the IC group significantly decreased (P〈0.05), whereas there were no significant differences on GPxl, GPx7 and CAT mRNA expression between NC and IC groups(P〉0.05) ;additionally,SOD1 and CAT mRNA expression in the pancreas of the IL group was
