摘 要:为探讨促乳素释放激素受体(prolactin releasing peptide receptor,PrRP-R)基因在不同发育时期绵羊睾丸和不同组织(心脏、肝脏、脾脏、肺脏、肾脏、背最长肌、股二头肌、下丘脑、垂体、松果腺)中表达的差异,以0,2,5,12,24月龄的小尾寒羊为研究对象,应用荧光实时定量PCR(quantitative real-time PCR)和免疫组化(immunohistochemistry)技术分别分析了PrRP-R基因在组织中的差异表达与定位。结果显示:PrRP-R基因在睾丸与心脏、肝脏、脾脏、肺脏、肾脏、背最长肌、股二头肌、下丘脑、垂体、松果腺之间的表达差异极显著(P<0.01),股二头肌、垂体、松果腺之间表达差异不显著(P>0.05),但背最长肌、下丘脑、垂体之间表达差异显著(P<0.05);0,2,5,12,24月龄的睾丸组织中均有一定量的表达,24月龄时表达最高(P<0.01);PrRP-R在0~24月龄绵羊睾丸组织中呈阶段性表达,0,2,5月龄时,可观察到PrRP-R在精原细胞中到阳性表达;12,24月龄时,在初级精母细胞和次级精母细胞中检测到强阳性表达。结果表明:PrRP-R在不同的组织和睾丸中表达并存在一定的差异,可能对绵羊的繁殖性能具有重要的调控作用。In order to explore the prolactin releasing peptide receptor(PrRP-R)gene expression difference between raw testicles and other tissues (heart, liver, spleen, lung, kidneY, longissimus dorsi,musculi biceps femoris, hypothalamus, pituitary and pineal gland), small tailed Han sheep were selected as the experiment objects. QRT-PCR and immunohistochemical technique were used to analyze the different expressions of the PrRP-R gene and localization in testicles. The results showed that PrRP-R gene had significant expression differences between testes and heart, liver, spleen, lung, kidney, longissimus dorsi, museuli biceps femoris, hypothalamus, pituitary, pineal glands (P〈0.01). and no significant expression differences(P〈0.05) between musculi biceps femoris, pituitary and pineal glands,but that of longissimus dorsi, hypothalamus and pituitary ex- pressed significant difference (P〈0.05). 0,2,5,12 and 24 months of testieular tissue had a cer- tain amount of express,the highest at 24 months (P〈0.01). PrRP-R was periodically expression in sheep testicular tissue from 0 to 24 months, positive expression of PrRP-R can be observed in spermatogonium of 0,2 and 5 months testicles. Strong positive expression of PrRP-R can be de- tected in the primary spermatocyte and secondary spermatocyte of 12 and 24 months testicles. SoPrRP-R expression in different organizations and testicles exists certain differences,may have an important regulatory effect on the reproduction performance of sheep.
