摘  要:目的:通过考察通络醒脑泡腾片干预后MID模型大鼠海马主要能量转运蛋白表达揭示其抗痴呆的作用机制。方法:建立微血栓诱发的MID大鼠模型,给予通络醒脑泡腾片干预90 d,采用Morris水迷宫评价大鼠的学习记忆功能,采用免疫组化和图像分析法评价大鼠海马GLUT-1、MCT-1和MCT-2的表达。结果:与模型对照组比较,通络醒脑泡腾片能显著缩短MID模型大鼠的逃避潜伏期,增加进入有效区域次数并延长有效区域停留时间,差异有统计学意义;通络醒脑泡腾片能显著促进模型大鼠海马GLUT-1、MCT-1和MCT-2表达。结论:通络醒脑泡腾片改善MID模型大鼠学习记忆功能的作用与其促进增强海马能量转运蛋白GLUT-1、MCT-1和MCT-2的表达有关。Objective: To study the effect of Tongluoxingnao effervescent tablet(TLXNET) on the expression of energy transport-related proteins in multi-infarct dementia(MID) rats in order to explore its mechanism of anti-dementia.Methods: Microthrombus was used to establish MID model rats which were intervened by gastric infusion of TLXNET for 90 d. Evalneting the learning and memory ability of MID model rats via Morris water maze. Additionally the expressions of GLUT-1,MCT-1 and MCT-2 in hippocampal were measured by immunohistochemical and image analysis. Results:Compared with the model control group,TLXNET could significantly reduce the escape latency,increase times of entrance into the valid region and prolong the residence time of MID model rats,which all revealed a significant statistic; meanwhile the expressions of GLUT-1,MCT-1 and MCT-2 were notably raised after administration of TLXNET,with an increase in the average optical density. Conclusion: TLXNET can improve learning and memory deficit which is related with promoting the expression of energy metabolism-related proteins transportation such as GLUT-1,MCT-1 and MCT-2.
