摘  要:利用均匀设计法对超临界CO_2流体提取桑叶黄酮的工艺进行优化,主要考察提取温度、提取压力、提取时间和夹带剂乙醇用量对桑叶黄酮提取率的影响.结果表明:超临界CO_2流体提取桑叶黄酮的最佳提取条件为提取温度51℃,提取压力36 MPa,提取时间4.0 h,夹带剂乙醇用量3.7 m L/g.实际测得的桑叶黄酮的提取率为1.42%,与模型预测值相符,说明采用均匀设计法优化超临界CO_2流体提取桑叶黄酮的工艺准确可靠.此外,与回流提取法、超声波辅助提取法相比,超临界CO_2提取法具有提取温度低、溶剂使用少、提取率相对较高的特点,适于桑叶黄酮提取.Uniform design methodology for the extraction process of flavonoids of mulberry leaves by super-critical CO2fluid(SCF) was studied. The effects of extraction temperature, extraction pressure, extraction time and the amount of ethanol entrainer on the yields of flavonoids were investigated. The results show that the optimum extraction conditions of flavonoids of mulberry leaves were extraction temperature 51 ℃, extraction pressure 36 MPa, extraction time 4.0 h, amount of ethanol entrainer 3.7 m L·g-1. The actual measured the yields of flavonoids is 1.42% and is basically consistent with the predicted value. These show that using uniform design methodology to model the extraction process of flavonoids of mulberry leaves by SCF was accurate and reliable. Additionally, compared with the reflux extraction and the ultrasonic-assisted extraction, SCF extraction achieved relatively acceptable extraction efficiency using less ethanol solvent at a lower extraction temperature, is a suitable method to extract flavonoids of mulberry leaves.
