摘  要:本研究利用DNA池和测序技术,结合PCR-RFLP方法,筛查了POU1F1基因第2外显子和第5内含子的SNP位点,分析了西藏申扎(SZ)、类乌奇(LWQ)、斯布(SB)和帕里(PL)4个牦牛类群(品种)共182个个体的POU1F1基因的多态性及其与体重、体高、体长等生长性状指标之间的相关性。结果表明:①POU1F1基因第2外显子高度保守,未发现任何突变,第5内含子中发现2个突变位点,分别为T727C(BglⅡ酶切)位点和T284C(StuⅠ酶切)位点。②申扎(SZ)、类鸟奇(LWQ)、斯布(SB)和帕里(PLl)4个群体中StuⅠ位点为中度多态性,其基因T/C频率分别为:0.61/0.39、0.54/0.46、0.54/0.46和0.62/0.38,BglⅡ位点为低度多态性;Hardy-Weinberg检验表明StuⅠ位点和BglⅡ位点均处于平衡状态(P>0.05)。③StuI位点在4个群体中表现CC、CT和TT 3种基因型,BglⅡ位点在4个群体中只表现CT和TT2种基因型。④帕里牦牛StuⅠ位点CC和CI基因型在生长性状上存在显著差异(P<0.05),其中CC基因型的体高和体长均高于CT基因型,而其他几个牦牛类群未发现类似现象。BglⅡ位点基因型与生长指标的相关性不显著。西藏牦牛POU1F1基因内含子5内T284C基因座表现的多态性可以作为改良其生长性状的一个遗传标记。This research on the use of DNA pool and sequencing technology, the PCR-RFLP method, was conducted to study the correlation of polymorphisms of POU1F1 gene and growth traits of Shenzha Yak, Leiwuqi Yak, Sibu Yak, Pall Yak by screening the SNP locus of gene exons 2 and intron 5 with three pairs of primer, a total of 182 yaks. The results showed that(i) no polymorphism was found at POU1F1 gene exon 2, which was highly conserved, two polymorphisms were found in intron 5, they were respectively T727C locus (Bgl Ⅱ) and T284C locus (Stul). (ii) In four populations of SZ, LWQ, PL, SB, StuI locus belonged to the middle polymorphic index, moreover, the frequencies of T/C alleles were respectively 0.61/0.39, 0. 54/0. 46 , 0. 54/0. 46 and 0.62/0.38, CT was a predominant genotype and T was a predominant allele. StuI locus belonged to the low polypeptide. StuI and Bgl Ⅱ locus were under Hardy-Weinberg equilibrium( P 〉0.05 ). (iii) StuI locus of four groups were found three genotype with CC, CT and TT. Only two genotypes( CT and TT) were found at BgllI locus. (iv)The correlation of polymorphisms between POUIF1 gene and growth traits among the four populations was analyzed. In Pall Yak, the results showed that Genotype CC and TT were highly associated with body length and body height( P 〈0.05 ), genotype CC was higher than that genotype CT, the same result did not appear in the left three population. The correlation between Bgl Ⅱ locus and growth index was not signif- icant. T284C locus polymorphism at POUIF1 intron 5 of Tibetan yak can be as a genetic marker for improved growth traits.
