摘 要:目的探讨吉西他滨对人食管癌Eca-109细胞的增殖、细胞周期、细胞凋亡及STAT3蛋白表达的影响。方法 MTT法测定吉西他滨对人食管癌Eca-109细胞的增殖抑制作用,PI单染流式细胞术检测人食管癌Eca-109细胞周期,Annexin V-FITC/PI双染流式细胞术检测人食管癌Eca-109细胞的凋亡情况,蛋白质印迹法检测STAT3蛋白的表达情况。结果不同浓度吉西他滨对人食管癌Eca-109细胞的抑制率均明显高于正常对照组(P<0.05),且随着药物浓度的增加,抑制率越高;吉西他滨0.20 mg/L组、0.40 mg/L组及0.80 mg/L组G0/G1期人食管癌Eca-109细胞百分数逐渐下降(P<0.05),G2/M期细胞百分数无变化(P>0.05),而S期细胞百分数逐渐增加(P<0.05)。吉西他滨0.20 mg/L组、0.40 mg/L组、0.80 mg/L组细胞凋亡率均明显高于正常对照组(P<0.05),且上述三组的STAT3蛋白表达均低于正常对照组(P<0.05),呈剂量依赖性。结论吉西他滨对人食管癌Eca-109细胞具有增殖抑制作用,阻滞细胞于S期,明显促进细胞凋亡,下调STAT3蛋白的表达。Objective To explore the effects of gemcitabine on cell proliferation, cell cycle, apoptosis and STAT3 protein expression in human esophageal cancer cell Eca-109. Method Inhibitory effect Of gemcitabine on proliferation of human esophageal cancer cell Eca-109 was detected by MTT method, cell cycle of human esophageal cancer cell Eca-109 was detected by PI single staining flow cytometry, apoptosis of human esophageal cancer cell Eca- 109 was detected by AnnexinV-FITC/PI AnnexinV-FITC / PI double staining flow cytometry. The expression of STAT3 protein was detected by Western blotting. Result The inhibition rate of different concentrations of gemcitabine on human esophageal cancer cell Eca-109 was significantly higher than that in normal control group (P〈 0.05), with the increase of drug concentration, the inhibition rate increased. The percentage of Eca-109 human esophageal cancer line G0/G1 in 0.20 mg/L group, 0.40 mg/L group and 0.80 mg/L group decreased gradually (P 〈 0.05), the percentage of G2/M cells was not changed (P 〉 0.05), the percentage of S cells increased gradually (P 〈0.05). The apoptosis rates of gemcitabine 0.20 mg/L group, 0.40 mg/L group and 0.80 mg/L group were significantly higher than those of normal control group (P〈 0.05), the expression of STAT3 protein in the three groups was lower than that in normal control group (P〈 0.05). Conclusion Gemcitabine can inhibit the proliferation of human esophageal cancer cell Eca-109, block cells in S phase, significantly promote apoptosis, down-regulated expression of STAT3.
