摘  要:目的:建立采用高效液相色谱同时测定维吾尔药阿纳其根中2个N-烷基类成分含量的方法,为制定阿纳其根质量标准提供依据。方法:采用LC-20A型高效液相色谱仪,Agilent TC-C_(18)色谱柱(4.6 mm×150 mm,5μm),以甲醇-水(70:30)为流动相,流速0.8 mL·min^(-1),检测波长243 nm,柱温25℃。结果:十-2E,4E-二烯酸-4-羟基苯乙胺和十二-2E,4E-二烯酸-4-羟基苯乙胺质量浓度分别在30.6~122.4 mg·L^(-1)和17.2~68.8 mg·L^(-1)范围内呈良好线性关系,平均加样回收率(n=6)分别为99.90%(RSD=1.60%)和100.4%(RSD=0.85%);3批阿纳其根药材中两者含量范围分别为0.046%~0.054%和0.035%~0.039%(n=3)。结论:经方法学验证,该方法可为阿纳其根药材质量控制提供一定的参考。Objective: To establish an HPLC method for the simultaneous determination of dodeca-2E, 4E-dienoic acid 4-hydroxyphenylethylamide and deca-2E, 4E-dienoic acid 4-hydroxyphenylethylamide in the roots from Anacyclus pyrethrum DC. , whichis a kind of Uyghur medicine. Methods: The analysis was conducted on a type of LC-20A HPLC, and was performed on a Agilent TC-C18( 4.6 mm× 150 mm, 5 μm ) column with mobile phase of methanol and water ( 70 : 30 ). The flow rate was 0.8 mL· min-1, the detection wavelength was 243 nm, and the column temperature was 25℃ The two effective components in 3 batches of medicines were analyzed. Results: The calibration curves of dodeca-2E, 4E-dienoic acid 4-hydroxyphenylethylamide and deca-2E, 4E-dienoic acid 4-hydroxyphenylethylamide were linear within the ranges of 30.6-122.4mg·L-1 and 17.2-68.8mg·L-1; the average recovery rates were 99.90% and 100.4% with RSDs of 1.60% and 0.85% ( n=6 ). The content of two components in 3 batches was in a ranges of 0.046%-0.054% and 0.035%-0.039% ( n=3 ), respectively. Conehlsion: The method is proved by methodology validation that it can provide some reference for the quality control of Anacyclus pyrethrum DC.
