摘 要:目的观察二氢杨梅素(dihydromyricetin,DHM)治疗溃疡性结肠炎(ulcerative colitis,UC)的效果并探讨其机制。方法 60只BALB/c小鼠采用随机数字表法分为正常组,模型组,DHM低剂量[50 mg/(kg·d)]、中剂量[100 mg/(kg·d)]、高剂量[200 mg/(kg·d)]组,5-氨基水杨酸[5-amino saliciylic acid,5-ASA,50 mg/(kg·d)]组。分组当日给予正常组小鼠蒸馏水自行饮用,其余组小鼠给予4%葡聚糖硫酸钠(DSS)水溶液7 d诱导急性UC模型。于饮用DSS当日开始,用DHM各剂量组及5-氨基水杨酸组按各组拟定剂量分别进行灌胃处理,连续给药14 d。对比各组疾病活动指数(DAI)、结肠长度、脾脏指数的变化、结肠组织学评分,肠组织TLR2、TLR4和NF-κB的活性以及炎性细胞因子IL-1β、TNF-α、IL-6表达及MPO、NO、SOD的活性变化。结果与模型组比较,DHM可显著降低结肠组织中TLR2、TLR4、NF-κB的表达,减少炎性因子如IL-1β、TNF-α、IL-6水平,同时还能够减少肠组织中MPO、NO活性及增加SOD的活性。其中DHM高剂量组优于低、中剂量组,差异有统计学意义(P<0.05),治疗效果最明显,其与5-氨基水杨酸组相比差异无统计学意义(P>0.05),治疗效果相当。结论 DHM可显著缓解DSS诱导的小鼠结肠炎症,其作用机制可能与抗炎、抗氧化有关。Objective To observe the curative effect of dihydromyricetin (DHM) in treating ulcerative colitis (UC) and to explore its mechanism. Methods Sixty BALB/c mice were randomly divided into normal group, model group, low-, middle-, and high-dosed DHM treatment groups and 5- ASA control group, with 10 mice in each group. The normal group was given distilled water to drink, and the other groups were given 4% dextran sodium sulfate (DSS) solution for 7 d to induce the acute UC model. At the beginning of the day of drinking DSS, the drugs were given to the mice of DHM and 5-ASA treatment groups respectively through intragastric continuous administration for 14 d. Disease activity index (DAI) , colon length, spleen index changes, histological scores, activities of TLR2, TLR4 and NF-kappa B, expression levels of inflammatory cytokines IL-1β, TNF-α, IL-6 and activities of MPO, NO, SOD in intestinal tissue were compared among the groups. Results Compared with model group, DHM could significantly reduce the expression of TLR2, TLR4, NF-KB in colon tissue, reduce the levels of inflammatory cytokines such as IL- lβ, TNF-α and IL-6, and also reduce the production of MPO and NO and increase SOD activity in intestinal tissue. The high-dosed treatment group had better results than low- and middle-dosed treatment groups, with significant with differences (P 〈 0. 05 ) , and the treatment effect was most obvious and was equivalent to the action of 5-ASA (P 〉 0. 05 ). Conclusion DHM can significantly alleviate the DSS induced colitis in mice,and its mechanism may be related to its anti-inflammatory and antioxidant activities.
