摘 要:目的探索硝酸沉淀蛋白与微波消解法对血中元素测定的影响。方法采用5%硝酸沉淀蛋白及微波消解法分别处理全血样,并保证最终硝酸浓度基本相同;标准曲线采用标准加入法,以消除基质干扰。结果各元素检出限,硝酸沉淀蛋白法为0.5~39μg/L,微波消解法为0.5~32μg/L;相关系数,硝酸沉淀蛋白法除硒以外,其余均≥0.998,微波消解法除铬以外,其余均≥0.998;精密度,硝酸沉淀蛋白法为1.4%~4.8%,微波消解法为4.1%~49.2%;加标回收率,硝酸沉淀蛋白法除铝外,其余均在82.5%~113%范围内,微波消解法在82.0%~123%范围内。结论血中As、Pb、Cd、Cr、Zn的测定前处理宜采用硝酸沉淀蛋白法,Se、Al宜采用微波消解法,而Mn、Cu则两种前处理方法均适用。Objective This study was to explore the effect of nitric acid precipitation of protein and microwave digestion on the determination of the elements in blood by ICP-MS. Methods The whole blood samples were pretreated using 5% nitric acid or microwave digestion to remove protein and to ensure that the final concentration of nitric acid was basically the same. The standard curves were prepared by standard addition method to eliminate the matrix interference. Results The detection limits were 0.5 -39 μg/L for the nitric acid pretreatment method and 0.5-32μg/L for the microwave digestion. For the removal of protein by nitric acid, the correlation coefficient (r) was ≥0.998 for the determination of all elements except Se. For the method of microwave digestion, r was ≥ 0.998 for all elements except for Cr. Relative standard deviations were 1.4%-4.8% for nitric acid pretreatment method and 4.1%-49.2% for microwave digestion.The recovery rates were 82.5%- 113% for nitric acid pretreatment method except for the determination of Al and 82.0%-123% for microwave digestion. Conclusion For the determination of As, Pb, Cd, Cr and Zn in the blood, samples should be pretreated with nitric acid method. For Se and Al, samples should be pretreated with microwave digestion method, and for Mn and Cu, both methods can be used.
