摘 要:目的探讨掺锶聚磷酸钙骨修复材料对成骨细胞血管内皮生长因子(VEGF)蛋白分泌量的影响。方法制备不同含锶量的掺锶聚磷酸钙骨修复材料,采用酶联免疫吸附(ELISA)法测定成骨细胞的VEGF蛋白分泌量,并采用噻唑蓝(MTT)法考察其对成骨细胞增殖的影响。结果 ELISA法结果表明,培养7 d后,1%,3%,5%,8%,10%含锶量的掺锶聚磷酸钙作用的成骨细胞VEGF分泌量均高于聚磷酸钙,差异均有统计学意义(t=6.561,11.454,11.256,15.149,9.778,P<0.05),其中,8%含锶量掺锶聚磷酸钙作用的成骨细胞VEGF分泌量最高。MTT法结果表明,培养3,5,7,9 d后,8%掺锶聚磷酸钙、聚磷酸钙浸提液作用的成骨细胞的吸光度(A)值高于阴性对照,差异均有统计学意义(t=3.960,5.029,5.229,4.808,2.584,2.954,2.657,2.798,P<0.05);培养5,7,9 d后,8%掺锶聚磷酸钙浸提液作用的成骨细胞的A值高于聚磷酸钙,差异均有统计学意义(t=2.583,2.930,1.934,P<0.05)。结论掺锶聚磷酸钙作为一种新型的骨修复材料,能促进成骨细胞分泌VEGF,具有良好的临床应用前景。Objective To learn the effect of strontium doped calcium phosphate bone repair material on VEGF protein secretion in osteoblasts and to provide a new bone repair material for orthopedics clinic. Methods The strontium doped calcium phosphate bone repair materials with different strontium content were prepared. The VEGF protein secretion was measured by ELISA method, and the effect of MTT on the proliferation of osteoblasts was investigated. Results The results of ELISA method after 7 d showed that the secretion of VEGF in osteoblasts of 1%, 3%, 5%, 8%, 10% strontium doped calcium phosphate were higher than calcium phosphate, and the differences were statistically significant(t =6.561, 11.454, 11.256, 15. 149, 9.778, P 〈 0.05).Among them, the content of VEGF was the highest in the 8% calcium containing calcium strontium calcium phosphate. The results of MTT method showed that the A values of 3, 5, 7, 9 d was higher than that of the negative control, and the value of the osteoblasts was higher than that of the negative control, and the differences were statistically significant( t = 3. 960, 5. 029, 5. 229, 4. 808, 2. 584, 2. 954, 2. 657, 2. 798, P 〈 0.05). After culture 5, 7, 9 d, the A value of the osteoblast of the 8% calcium doped calcium phosphate solution were higher than that of calcium phosphate, and the differences were statistically significant( t =2. 583, 2. 930, 1. 934, P 〈 0.05). Conclusion As a new bone repair material, strontium doped calcium phosphate can promote the secretion of VEGF, which has a good clinical application prospects.
