摘 要:为了研究奶牛黄体的形成及退化过程的相关机理,并进一步探究更加准确的妊娠及相关生理过程的蛋白标记物,试验采用蛋白质组学经典的二维凝胶电泳(2-DE)技术检测和分析妊娠前后黄体组织间的差异蛋白,使用PDQuest8.0软件检测,取差异蛋白点进行分析,并经MALDI TOF/TOFMS/MS检测。结果表明:6种差异蛋白分别为26s蛋白酶体蛋白、纤维蛋白、DNA拓扑异构酶蛋白、核纤层蛋白、硫氧还原蛋白、微管蛋白。对这6种蛋白在组织间的表达差异进行分析,发现这些蛋白可能与黄体形成和黄体退化及黄体激素分泌有关。To study the related mechanisms of formation and degradation processes of corpus luteum in dairy cattle, and further explore the more accurate protein markers related to pregnancy and related physiological processes, two - dimensional gel electrophoresis (2 - DE) as a classical method in proteomics was used to detect and analyze the differential proteins between the luteal tissues before and after pregnancy. The differen- tial protein spots were analyzed by PDQuest8.0 software and detected by MALDI TOF / TOF - MS / MS, respectively. The results showed that six kinds of differential proteins were 26s preteasome protein, DNA topoisomerase protein fibrin, DNA topoisomerase protein topoisomerase, lamin, thioredoxin, and tubulin, respectively. The differences in the expressions of the six proteins between the tissues were analyzed, and it was found that these proteins might be associated with the formation and degradation of the corpus luteum and luteal degradation, as well as the secretion of luteal hormones.
