摘 要:为了探究转pGH、IGF-1双基因猪F1代阳性率及表达差异情况,试验采用PCR、测序及荧光定量PCR检测转pGH、IGF-1双基因猪的mRNA表达水平。结果表明:53头F1代猪中17头为阳性猪,阳性率为32.08%,证实外源pGH和IGF-1基因已遗传到F1代中;1月龄到4月龄转双基因猪与非转基因猪IGF-1和pGH基因平均表达量均无显著性差异(P>0.05);5月龄转双基因猪pGH和IGF-1基因平均表达量分别是非转基因猪的1.98倍和2.00倍,存在显著差异(P<0.05);6月龄转双基因猪pGH和IGF-1基因平均表达量分别是非转基因猪的1.67倍和1.73倍(P<0.05)。说明pGH、IGF-1双基因已遗传至F1代猪中,并且能够在猪体内稳定表达。To explore the positive rate and expression differences of the F1 generation of genetically modified pigs with the double genes of pGH and IGF - 1, the methods of PCR detection, sequencing and fluorescence quantitative PCR were used to detect the mRNA expression levels of genetically modified pigs with the double genes of pGH and IGF - 1. The results showed that 17 positive pigs were found in 53 pigs of the F1 generation, and the positive rate was 32.08%, which was confirmed that the pGH and IGF - 1 genes had been inherited to the F1 generation. There were no significant differences ( P 〉 0.05 ) in the average expression levels of the pGH and IGF - 1 genes between the genetically modified pigs and non - genetically modified pigs from one month to four months of age, but the average expression levels of pGH and IGF - 1 genes in the genetically modified pigs with the double genes were 1.98 times and 2. (30 times higher than those in the non - genetically modified pigs at five months of age, respectively, while the difference were significant( P 〈 0. 05 ). The average expression levels of the pGH and IGF - 1 genes in the genetically modified pigs with the double genes were 1.67 times and 1.73 times higher than those in the non - genetically modified pigs at six month of age, respectively, while the differences were significant ( P 〈 0. 05 ). The results indicate that the double genes of pGH and IGF - 1 have been inherited to the pigs of the F1 generation and can be stably expressed in pigs.
