复荣通脉胶囊对糖尿病大鼠胸主动脉PAI-1、t-PA表达的影响

摘　　要：目的观察复荣通脉胶囊对糖尿病大鼠胸主动脉1型纤溶酶原激活物抑制剂(PAI-1)、组织型纤溶酶原激活物(t-PA)表达的影响,探讨其对纤溶系统的作用。方法将Wistar大鼠随机分为对照组和糖尿病造模组,造模成功大鼠随机分为模型组、复荣通脉低剂量组(0.7 g/kg)、复荣通脉中剂量组(1.4 g/kg)、复荣通脉高剂量组(2.8 g/kg),8周后,观察各组大鼠血糖、血脂水平及胸主动脉PAI-1及t-PA免疫组化表达。结果造模组大鼠血糖较对照组明显升高(P<0.05);胸主动脉PAI-1表达在各造模组较对照组明显增强(P<0.05),而在复荣通脉中剂量组、高剂量组表达明显受抑制;胸主动脉t-PA表达在各造模组较对照组明显减弱(P<0.05),在复荣通脉中剂量组、高剂量组其表达明显增强(P<0.05)。结论复荣通脉胶囊可抑制PAI-1、促进t-PA在大鼠胸主动脉的表达。Objective To observe the effects of Furong Tongmai capsule（ FTC） on the expressions of plasminogen activator inhibitor-1（ PAI-1）and tissue plasminogen activator（ t-PA） in thoracic aorta tissue of diabetic rats,and explore its possible mechanism. Methods Wistar rats were randomly chosen as blank control group and diabetes model group. The diabetic model rats were randomly divided into DM,FTC low-dose group,FTC medium-dose group,FTC high-dose group. Aftet 8 weeks later,we observed the blood sugar and the expression levels of PAI-1,tPA with immunohistochemical technique. Results The blood sugar of diabetic rats was significantly higher than that in control group. The expression of PAI-1 in thoracic aortic was decreased in FTC medium-dose group and FTC high-dose group. The expression of t-PA in thoracic aortic was increased in FTC medium-dose group and FTC high-dose group. Conclusion FTC could improve the expression of t-PA,inhibit the over-expression of PAI-1 in thoracic aorta tissues of diabetic rats.
