摘  要:目的探讨深低温停循环对大鼠大脑信号转导分子4(brain signal transduction molecule 4,Smad4)和Nestin蛋白表达的影响。方法 45只SD大鼠随机分为深低温停循环组、常温停循环组和常温不停循环组各15只,建立体外循环后3组大鼠肛温分别控制在<20.0℃、36.5~37.0℃、36.5~37.0℃,深低温停循环组和常温停循环组大鼠体外循环10min后停循环10min,常温不停循环组大鼠体外循环20min后处死大鼠取额顶叶皮质脑组织行组织病理学HE染色,采用Western blot法检测大鼠脑组织Smad4和Nestin蛋白表达情况。结果常温停循环组大鼠大脑呈轻度缺血性改变,深低温停循环组和常温不停循环组大鼠大脑椎体细胞核仁清楚,呈椭圆形;深低温停循环组大鼠脑组织中Smad4和Nestin呈高水平表达,常温停循环组大鼠脑组织中Smad4和Nestin呈中等水平表达,常温不停循环组大鼠脑组织中Smad4和Nestin呈少量表达;深低温停循环组大鼠大脑组织Smad4(105.23±8.04)和Nestin(103.98±4.16)平均灰度值明显低于常温停循环组(118.38±7.24、120.52±6.73)和常温不停循环组(136.85±5.78、140.41±6.27)(P<0.05),常温停循环组明显低于常温不停循环组(P<0.05)。结论停循环可上调大鼠脑组织中Smad4和Nestin的表达水平,深低温可进一步促进停循环脑组织中Smad4和Nestin的表达水平。Objective To explore the influence of deep hypothermic circulatory arrest on the expression of brain signal transduction molecule 4 (Smad4) and Nestin protein in rats. Methods Forty-five SD rats were randomly divided into deep hypotherrnic circulatory arrest group, room-temperature circulatory arrest group and room-temperature cyclic group, with 15 rats in each group, keeping the rectal temperature at 〈20, 36. 5 to 37 and 36. 5 to 37 ℃, respectively. Deep hypothermic circulatory arrest group and room-temperature circulatory arrest group received cardiopulmonary bypass for 10 min followed by circulatory arrest for 10 min. Room-temperature cyclic group received cardiopulmonary bypass for 20 rain at room temperature without circulatory arrest. All groups underwent HE staining, and the expressions of Smad4 and Nestin protein were detected by Western blot method. Results Room-temperature circulatory arrest group had mild cerebral ischemic changes, and deep hypothermic circulatory arrest group and room-temperature cyclic group were found clear and oval cerebral vertebral cells nucleolus. Deep hypothermic circulatory arrest group had high levels of Smad4 and Nestin protein, room-temperature circulatory arrest group had medium levels of Smad4 and Nestin protein, and room-temperature cyclic group had low levels of Smad4 and Nestin protein. The average gray values of Smad4 and Nestin protein were significantly lower in deep hypothermic circulatory arrest group (105.23±8.04, 103.98± 4.16) than those in room-temperature circulatory arrest group (118. 38 ±7. 24, 120. 52±6. 73) and room-temperature cyclic group (136.85±5.78, 140.41±6.27) (P〈0.05), and were significantly lower in room-temperature circulatory arrest group than those in room-temperature cyclic group and Nestin protein, and deep hypothermia arrest (P〈0. 05).
