摘  要:目的:研究枸杞总黄酮提取物(TFLB)对链脲佐菌素(STZ)诱导建立的糖尿病(DM)大鼠损伤模型的保护作用及其可能的分子机制。方法:采用STZ诱导建立DM大鼠损伤模型,实验分为正常对照组、DM模型组和TFLB干预组(60,90,120mg·mL^(-1))。正常对照组及DM模型组大鼠每天每只予以等量生理盐水灌胃,干预组大鼠以不同浓度TFLB灌胃4周。灌胃满4周后,以尾静脉采血测空腹血糖,胸腔采血测定血脂,摘取胰腺组织做HE染色和免疫组化。结果:与正常对照组相比较,DM模型组大鼠血糖、血脂显著升高(P<0.05,P<0.01);胰岛形态皱缩,体积变小,状态不规则,胰岛数量及岛内分泌细胞明显减少,且细胞稀疏不均排列紊乱。与DM模型组相比,TFLB的干预可以延缓、延迟血糖升高的进程,对血脂有明显降低趋势,具有显著性差异(P<0.05,P<0.01),随TFLB剂量的升高,大鼠胰岛数量及岛内分泌细胞数有所增多,细胞排列相对均匀规整,胰岛结构、形态基本趋于正常,细胞空泡变性相对较少。结论:TFLB可以延缓血糖升高速度,调节血脂代谢的紊乱,进而推迟DM病程的进展,能明显保护DM大鼠的胰岛功能,改善大鼠的胰岛素抵抗。OBJECTIVE To investigate the protective effects and possible molecular mechanism of total flavonoids from Lycium barbarum(TFLB)on rats with diabetes mellitus(DM)induced by streptozotocin(STZ).METHODS DM rat models were established by using STZ,and randomly divided into three groups:normal control group,DM group and TFLB group.TFLB group was further divided into 60,90,120 mg·mL^-1 subgroups.Rats were gavaged equal volume of normal saline in normal control and DM group,and gavaged TFLB in TFLB group for 4 weeks.Four weeks later,blood was collected from tail vein to determine fast glucose,from thoracic cavity to determine blood fat.Pancreatic tissues were collected and stained by HE method for immunohistochemistry.RESULTS Compared with normal control group,rats in DM group had obviously increased blood glucose and fat(P〈0.05,P〈0.01),pancreatic islets shrank to smaller volumes and irregular shapes,islets and endocrine cells decreased significantly,and still tended to decrease obviously.Compared with DM group,TFLB delayed blood glucose increase,and had obvious declining effects to blood fat(P〈0.05,P〈0.01).As TFLB dose increased,pancreatic islets and endocrine cells increased,cells arranged relatively regularly,pancreatic structure and shape tended to be normal,cell vacuole degeneration relatively decreased.CONCLUSION TFLB can postpone blood glucose increase,adjust blood fat metabolism,postpone DM course,obviously protect pancreatic functions of DM rats,and alleviate insulin resistance.
