摘 要:采用SRAP标记对色素腺体性状不同棉花近等基因系进行分析。结果表明,39对SRAP引物组合对2对近等基因系材料共扩增出304条带,每对引物组合可以扩增出5.69条多态性带,多态性条带比率达到73%;共扩增出57条特异条带,特异条带比例达到18.9%。遗传相似度分析结果表明,W1与Y1遗传相似系数为0.740 3;Y2与W2的遗传相似系数为0.763 6。聚类分析结果表明,在GS为0.85处,将参试材料分为2个类群,W1与Y1的遗传相似系数比较大,归为一类;W2与Y2的亲缘关系比较近,归为另一类。研究结果可为近等基因系亲缘关系的鉴定以及为腺体基因的进一步研究提供依据。Near-isogenic lines with different pigment glands of cotton were analyzed by SRAP marker in this study. The result showed that a total of 304 bands were amplified by 39 SRAP primers combinations, 5.69 bands with polymorphism were amplified by per primer combination. The ratio of polymorphic bands was 73%, 57 bands(18.9%)were specific bands. The analysis of genetic similarity coefficients showed that the genetic similarity coefficient was 0.740 3 between W1 and Y1, the genetic similarity coefficient was 0.763 6between W2 and Y2. The cluster analysis showed that the materials were divided into two groups at the similarity coefficient of 0.85, W1 and W2 were clusted firstly, because their genetic similarity was bigger, W2 and Y2 were clusted another group, because their genetic relationship was relative close. The result will provide evidence for the identification of genetic relationship from near-isogenic lines and further study of pigment gland gene.
