摘 要:目的探讨脂联素(APN)及其基因位点+45、+276和-11377的单核苷酸多态性与广西壮族代谢综合征(MS)的相关性。方法采用酶联免疫吸附试验(ELISA)检测壮族MS组、壮族健康组、汉族MS组和汉族健康组(各100例)血清中APN浓度,聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测APN基因位点+45T/G、+276G/T和-11377C/G的多态性。结果 (1)较同民族健康组,两个民族的MS组高密度脂蛋白胆固醇(HDL-C)和APN均显著降低,而BMI、空腹血糖(FPG)、收缩压、舒张压、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)明显升高,差异均有统计学意义(P<0.05);(2)壮族、汉族MS组APN+45基因位点的TG和GG基因型频率明显比同族健康组升高,差异有统计学意义(χ2=14.319,P=0.001;χ2=11.631,P=0.003)。壮族、汉族MS组的APN+276位点TG和GG基因型明显高于同族健康组,差异有统计学意义(χ2=25.058,P=0.000;χ2=25.425,P=0.003)。4组的APN-11377位点绝大部分是野生型,虽等位基因分布频率略有差异,但差异均无统计学意义(P>0.05);(3)两个MS组中APN+45位点为TT基因型的患者血清APN水平显著高于基因型为TG和GG型的患者,差异有统计学意义(P<0.05)。APN+276位点为GG型的患者血清APN水平明显高于基因型为GT和TT型的患者,差异有统计学意义(P<0.05)。结论 APN+45位点G基因型和+276位点T基因型导致血清中APN水平降低可能与广西地区壮族和汉族MS发生有相关性。Objective To study the correlation between adiponectin(APN)and gene site APN+45,+276and-11377 polymorphisms with Zhuang metabolic syndrome(MS)in Guangxi Region.Methods The levels of serum adiponectin in Zhuang MS patients,Han MS patients,Zhuang normal people and Han normal people(each of 100cases)were detected by enzyme-linked immunosorbent assay(ELISA)method,and APN+45,APN+276and APN-11377 polymorphisms were detected by using PCRRFLP.Results(1)Compared with the same nationality normal control group,the APN and HDL-C levels of the two MS groups were significantly decreased,while BMI,FPG,systolic blood pressure,diastolic blood pressure,TC,TG and LDL-C were significantly increased,the differences were statistically significant(P〈0.05);(2)the TG and GG genotype frequency of APN+45loci in Zhuang and Han MS groups was significantly higher than that in the same nationality normal group,the differences were statistically significant(χ2=25.058,P=0.000;χ2=25.425,P=0.003).APN+276loci TG and GG genotype frequency in Zhuang and Han MS groups significantly higher than that of same nationality normal group,the differences were statistically significant(χ2=14.319,P=0.001;χ2=11.631,P=0.003).The heterozygosis degree of APN+276locus in the Zhuang and Han MS groups was significantly higher than that in the same nationality individuals,the difference was statistically significant(all P〈0.05).Most of APN-11377 locus in the 4groups was the wild type,although there was a slight difference in the distribution of allele frequencies,but the difference was not statistically significant(P〉0.05).(3)The serum APN level in the patients with APN+45TT genotype of the two MS groups was significantly higher than that in the patients with TG and GG genotype,and the differences were statistically significant(P〈0.05).The serum APN level in the patients with APN+276GG genotype was significantly higher than that in the patients with GT and TT genotype,and the differences
