摘  要:目的观察感染携带肝细胞生长因子的重组腺病毒(Ad-HGF)对高糖诱导的原代人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)一氧化氮(NO)生成的影响,并探究其机制。方法将HUVECs分为低糖组(LG组)、高糖组(HG组)、腺病毒对照组(HG+Ad-GFP组)和实验组(HG+Ad-HGF组),感染48 h后,采用NO荧光探针检测细胞内NO水平,蛋白免疫印迹法(Westren Blot)分别检测各组细胞鞘氨醇激酶1(sphingosine kinase1,SPHK1)、沉默信息调节因子2相关酶类1(silent mating type information regulation 2 homolog 1,SIRT1)和内皮型一氧化氮合酶(endothelial ntric oxide synthase,e NOS)蛋白表达。结果 HG组HUVECs细胞内NO水平及SIRT1和e NOS蛋白表达水平低于LG组,HG+Ad-HGF组高于HG组(P<0.05),但4组间SPHK1表达水平比较差异无统计学意义(P>0.05)。结论 Ad-HGF可通过激活SIRT1-e NOS-NO通路保护HUVECs免受高糖损伤。Objective To observe effect of adenovirushepatocyte growth factor (Ad-HGF) on production of nitric oxide (NO) in high glucose (HG) induced human umbilical vein endothelial cells (HUVECs) and its mechanism. Methods HUVECs were divided into low glucose (LG) , HG, HG transfect with Ad-HGF ( HG + Ad- HGF) and HG transfect with Ad- GFP (HG + Ad-GFP) groups. After infection for 48 h, NO levels were detected using NO fluorescent probe; Westren Blot method was used to detect expressions of sphingosine kinase 1 (SPHK1) , silent mating type information regulation 2 homolog 1 (SIRT1) and endothelial nitric oxide synthase (eNOS). Results NO levels and SIRT1 and eNOS expressions in HUVECs in HG group were lower than those in LG group, while the above values in HG + Ad- HGF group were higher than those in HG group (P 〈 0.05) , but there were no significant differences in SPHK1 expressions among the 4 groups (P 〉 0.05). Conclusion Ad-HGF can protect HUVECs from high glucose damage by activating SIRTI-eNOS-NO pathway.
