摘  要:目的探讨高糖条件下感染携带肝细胞生长因子的重组腺病毒(Ad-HGF)对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)凋亡相关蛋白Bax、Bcl-2表达的影响。方法将HUVECs分为低糖组(LG组,5.5 mmol/L)、高糖组(HG组,35 mmol/L)、腺病毒对照组(HG+Ad-GFP组)和实验组(HG+Ad-HGF组)。检测4组HUVECs增殖情况、细胞内活性氧(ROS)水平及凋亡相关蛋白Bax、Bcl-2的表达。结果 HG组、HG+Ad-GFP组HUVECs存活率低于LG组,HG+Ad-HGF组高于HG组(P<0.05,P<0.01)。HG组、HG+Ad-HGF组HUVECs内ROS水平高于LG组,但HG+Ad-HGF组低于HG组(P<0.05)。HG组、HG+Ad-GFP组Bax、Bax/Bcl-2高于LG组,Bcl-2低于LG组,但HG+Ad-HGF组Bax、Bax/Bcl-2低于HG组,Bcl-2高于HG组(P<0.05)。结论感染Ad-HGF可通过降低细胞内ROS水平和Bax/Bcl-2减少细胞凋亡,进而对高糖诱导的HUVECs起到保护作用。Objective To investigate effects of high glucose (HG) induced human umbilical vein endothelial cells (HUVECs) infected by adenovirushepatocyte growth factor (Ad-HGF) on protein expressions of apoptosis related Bax and Bcl-2. Methods HUVECs were divided into low glucose (LG, 5.5mmol/L), HG (35mmool/L) , HG transfeet with Ad-HGF ( HG + Ad- HGF) and HG transfect with Ad- GFP ( HG + Ad-GFP) groups. Proliferation conditions of HUVECs, reactive oxygen species (ROS) levels and expressions of apoptosis related proteins Bax and Bcl-2 were detec- ted. Results Survival rates of HUVECs in HG ancl HG + Ad-GFP groups, were lower than that in LG group, while the rate in HG+Ad-GFP group was higher than that in HG group (P〈0.05, P〈0.01). ROS ]eveisin HG and HG+Ad- HGF groups were higher than that in LG group, while the level in HG + Ad-HGF group was lower than that in HG group (P 〈 0. 05 ). Bax and Bax/Bcl-2 expressions were higher, while Bcl-2 expressions were lower in HG and HG + Ad-GFP groups than those in LG group; Bax and Bax/Bcl-2 values in HG + Ad-HGF group were lower than those in HG group (P 〈 O. 05). Conclusion Transfecting the Ad-HGF can achieve protection for HG induced HUVECs by decreasing the intracellular ROS level and Bax/Bcl-2 apoptosis rate.
