摘  要:目的:研究地格达-4味汤对"幽门结扎"诱导的肝损伤的作用。方法:取SD大鼠50只,分成5组,即正常、模型、地格达-4味汤高、中、低(166.4、83.2、41.6 g·kg-1;生药量分别为557、278.6、139.3 g·kg-1)剂量组,共给药15 d,末次给药后禁食不禁水24 h,除正常以外腹腔注射10%水合氯醛麻醉,开腹幽门结扎,关腹后单笼饲养18 h,腹主动脉取血,取肝脏,测定相关指标。结果:模型组与正常组比较血清AST、ALT量明显增加(P<0.01或P<0.05),肝组织GSH-PX明显降低(P<0.01);TBIL、DBIL、ALP明显增高(P<0.01);形态学观察肝细胞有明显的脂肪变、水肿、边膜外肝细胞大量坏死;肝细胞膜蛋白MRP3表达上调至3.5倍、MRP4表达下调至0.05倍、核受体Car下调至2.8倍、核受体Pxr下调至0.0019倍。各给药组与模型组比较地格达-4味汤中、高剂量组AST、ALT明显降低,GSH-PX明显增高(P<0.01);对TBIL、DBIL、ALP中剂量组明显降低(P<0.05);肝组织形态有一定量的好转;中剂量组膜蛋白MRP3下调至0.3倍、MRP4上调至2.7倍、核受体Car上调至3066倍、Pxr上调至506倍。结论:模型建立成功,地格达-4味汤具有明显的保肝作用,且它的作用机制为对核受体Car、Pxr的作用,而调节MRP3、MRP4表达发生改变。据结果分析中剂量组效果最佳。Objective: To study the effect of Mongolian Medicine "Digda -4 Decoction" on the acute liver injury of rats caused by pylorus ligation. Method: Fifty SD rats were divided into 5 groups,which were normal, model, Mongolian medicine "Digda-4 Decoction" high,medium and low (166.4,83.2,41.6 g·kg^-1) dose groups,a total of 15 days. At the end of the day, no allowance for food or water for 24 hour. On the next day morning, under normal abdominal injection of chloral hydrate anesthesia, open pyloric ligation. After operation, each rat was caged singly for 18 h and then the blood simple was taken out from abdominal aorta and liver, determining the related indicators. Results: Compared with normal group, serum AST and ALT in the model group were increased significantly ( P 〈 0.01 or P 〈 0. 05 ) and liver GSH - PH decreased obviously(P 〈 0. 01 ). TBIL, DBIL and ALP were significantly higher( P 〈 0.01). By macroscopic view, the observation model group rats had liver congestion and peritoneal effusion. Morphological observation showed hepatocyte obvious fatty change, edema and edge membrane cells necrosis. The expression level of MRP3 in liver cell membrane protein increased to 3.5 times and MRP4 expression level decreased to 0.05 times and nuclear receptor CAR decreased to 2. 8 times,nuclear receptor PXR decreased to 0. 001 9 times. Compared with model group, serum AST and ALT in the each dosage group were lower, especially those in high dose Digda -4 Decoction group was obviously lower and GSH - PX increased obviously (P 〈0.01 ). TBIL, DBIL and ALP in middle dose group decreased obviously( P 〈 0. 05 ) and there was a certain amount of improvement in liver tissue form. The medium dose group' s membrane protein MRP3 decreased to 0.3 times, MRP4 increased to 2.7 times and nuclear receptor CAR increased to 3066 times, PXR decreased to 506 times.
