摘 要:目的:探讨冬虫夏草提取物对结肠癌细胞株HT-29细胞增殖、凋亡的影响及其机制。方法:利用不同浓度冬虫夏草提取物(0.5~6.5 mg/m L)作用HT-29细胞24、48和72 h,采用MTT、平板实验、流式细胞检测和WB检测冬虫夏草提取物对HT-29细胞生长增殖作用、细胞集落数的形成、对细胞凋亡率的影响以及对细胞凋亡相关分子Caspase-3蛋白表达的影响。结果:MTT结果表明,提取物能够显著抑制HT-29的生长增殖,呈一定的量时效关系,半数抑制浓度为3.5 mg/m L;流式细胞仪分析表明,冬虫夏草提取物呈浓度依赖性诱导HT-29细胞凋亡,浓度为6.5 mg/m L时,HT-29凋亡率(12.57±0.16)%;Western blotting表明,HT-29细胞与对照组相比,经不同浓度提取物诱导后,Caspase-3表达逐渐增强。结论:冬虫夏草提取物具有抑制HT-29细胞增殖作用,这可能与增加Caspase-3蛋白表达、诱导细胞调亡有关。Objective: To investigate the effect of Cordyceps sinensis extracts on the proliferation and apoptosis of HT - 29 cells and the relevant molecular mechanism. Methods: The anti - proliferative effect of different concentrations of Cordyceps sinensis on HT - 29 was measured by MTT assay. Colony - forming capacity was measured by trypan blue staining. The apoptosis ratio of HT - 29 was analyzed by flow cytometry. The expression level of Casppase - 3 was analyzed by Western blot. Results :The MTT assay showed that Cordyceps sinensis had potent anti -proliferative effect on HT- 29 ceils in time and dose dependent manner. ICs0 was 3.5 mg/mL. Flow cytometry analysis revealed that Cordyceps sinensis increased the percentage of apoptotic HT- 29 cells and the apoptosis rate was (12.5 + 0.16)% when the concentration was 6.5 mg/mL. The expression level of Caspase - 3 was increased significantly in HT - 29 cells treated with different concentrations of Cordyceps sinensis. Conclusion : Cordyceps sinensis inhibits the proliferation and induces apoptosis of HT -29 cells by increasing Caspase- 3 expression.
