摘 要:目的:观察女贞子提取物(extract of nuzhenzi,ENZZ)对糖尿病大鼠早期肝损伤的干预作用及其机制。方法:采用饲喂高糖高脂饲料结合注射链脲佐菌素(streptozotocin,STZ)45 mg·kg-1的方法制作糖尿病大鼠的模型。造模成功大鼠随机分为模型组、ENZZ高、中、低剂量组(0.8、0.4、0.2 g·kg-1·d-1)、二甲双胍组(10 mg·kg-1·d-1),另取8只作为正常组灌喂4周,末次给药后处死大鼠,股动脉取血测空腹血糖(FBG)、血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、肝组织匀浆超氧化物歧酶(SOD)和丙二醛(MDA);取肝脏称重,计算肝脏指数,HE染色观察肝脏病理学形态变化。结果:与正常组相比较,模型组大鼠血糖、ALT、AST、肝组织匀浆MDA水平和SOD活性均有显著性差异(P<0.01)。与模型组相比较,ENZZ中、高剂量组大鼠血糖水平显著降低(P<0.01或P<0.05),肝脏指数下降(P<0.05),肝组织MDA水平显著降低(P<0.01或P<0.05),SOD活性提高(P<0.01或P<0.05),光镜下可见肝细胞病理改变(水肿、点状或灶状坏死及炎症细胞浸润)均有不同程度的改善,且降低程度与灌胃剂量具有正相关性;二甲双胍组大鼠空腹血糖显著下降(P<0.01)其他指标无显著性差异(P>0.05)。结论:女贞子提取物可以增强糖尿病大鼠肝脏的抗氧化防御功能,减轻糖尿病大鼠早期肝脏组织的损伤,具有一定的护肝作用。Objective: To observe the effect of extract of Nuzhenzi(ENZZ) intervention effects on liver injury in early diabetic rats and its mechanism. Methods: The diabetic rat models were induced by high fat diet with intraperitoneal injection of streptozotocin (STZ)45 mg kg-i. And then the model rats were randomly divided into model group, high, medi- um and low(0.8,0.4,0.2 g·kg^-1·d^-1) doses of ENZZ groups,mefformin group ( 10 mg·kg^-1·d^-1). At the same time, the other 8 rats of normal control group were fed 4 weeks. The rats were sacrificed after 4 weeks and blood samples were collected from each rat to test the fasting blood glucose (FBG) , serum alanine aminotransferase (ALT), aspartic acid transaminase ( AST), superoxide dismutase (SOD) and malondialdehyde (MDA) of liver tissue. Liver weight and the liver index were calculated. The pathological changes of the liver were evaluated with HE. Results : Compared with the normal group, the levels of blood glucose, ALT, AST, MDA level and SOD in liver tissue of model group were significantly different(P 〈 0. 01 ). Compared with model group, the levels of blood glucose were all significantly decreased in MENZZ and HENZZ groups ( P 〈 0. 01 or P 〈 0.05 ). The liver index decreased ( P 〈 0. 05 ) and MDA levels of liver tissue de- creased significantly ( P 〈 0. 01 or P 〈0. 05). The activity of SOD significantly increased ( P 〈 0. 05 or P 〈 0. 05 ) and the pathological change of liver ceil under light microscope ( edema, punetate or focal necrosis and inflammatory cell infil- tration) showed different degrees of improvement. The levels of blood glucose of metformin group decreased significantly ( P 〈 0. 01 ) and other indexes had no difference ( P 〉 0. 05 ). Conclusion : Extract of Nuzhenzi can enhance the antioxi- dant defense function and relieve the damage of early liver tissue in the liver of diabetic rats and it has the protective effect on liver injury at early stage in di
