摘 要:目的研究柴胡疏肝散对肝郁证大鼠行为学和大鼠海马及下丘脑单胺类神经递质的影响。方法用慢性束缚应激结合孤养法建立肝郁证大鼠模型,测评开野、蔗糖水实验,采用高效液相色谱法测定大鼠海马及下丘脑单胺类神经递质。结果模型组大鼠的行为学发生了明显变化,海马及下丘脑中5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、多巴胺(DA)含量均下降,去甲肾上腺素(NE)含量呈增加趋势,而柴胡疏肝散能调控其变化趋势。结论柴胡疏肝散能改善肝郁证大鼠行为学的变化,具有治疗肝郁证的作用,其作用机制可能与其能够调节肝郁证大鼠脑5-HT、5-HIAA、NE、DA有关,从而有效发挥抗肝郁作用。Objective To study the effects of Chaihu Shugan powder (CSD) on the ethological changes and levels of monoamine neu- rotransmitters in the hippocampus and hypothalamus in rats with liver- qi stagnation syndrome.Methods The rat model with liver-qi stagnation syndrome was established by chronic restraint stress and keeping them alone.Each rat was reviewed by Open - field test, Sucrose test and determined the levels of monoamine neurotransmitters in the hippocampus and hypothalamus by high performance liquid chromatography (HPLC).Results The ethological manifestation of model rats was changed obviously, and the levels of 5 - hydroxytryptamine (5- HT), 5- hydroxyindoleacetic acid (5 - HIAA),dopamine(DA) in the hippocampus and hypothalamus were decreased,while the level of norepinephrine (NE) was increased.CSD could regulate its trends.Conclusion CSD can improve the model rat's ethological manifestation and have therapeutic effects of liver -qi stagnation syndrome.The mechanism may be adjust the levels of 5 - HT,5 - HIAA, NE, DA in rat brain with liver - qi stagnation syndrome, in order to effectively play the role of anti - liver - qi stagnation.
