摘 要:目的探讨黄芪-葛根药对对吲哚美辛诱导的大鼠胃黏膜损伤的保护作用。方法取SD大鼠,按体质量分层随机分为正常组,模型组,阳性药组(西咪替丁0.12 g·kg^(-1)),药对高、中、低剂量组(黄芪-葛根药对水煎液24,12,6 g·kg^(-1)),连续灌胃4 d,第4天给药后75 min,除正常组外,其余各组均腹腔注射吲哚美辛混悬液35 mg·kg^(-1)造模,观察胃黏膜损伤分数、胃黏膜病理学改变,检测胃黏膜组织丙二醛(MDA)、超氧化物歧化酶(SOD)、活性氧自由基(ROS)的水平。结果模型组大鼠胃黏膜出血、糜烂、损伤明显,胃黏膜组织ROS、MDA含量上升,SOD活力下降。与模型组比较,黄芪-葛根药对各剂量组能不同程度地改善大鼠胃黏膜外观及病理学的变化,降低胃黏膜损伤分数(P<0.05,P<0.01);同时,明显减少大鼠胃黏膜组织MDA、ROS含量(P<0.05,P<0.01);黄芪-葛根药对高剂量组能明显提高SOD活力(P<0.05)。结论黄芪-葛根药对对吲哚美辛诱导的大鼠胃黏膜损伤有一定保护作用,其作用可能与抗胃黏膜组织氧化应激水平有关。Objective To explore the protective effect of drug pair Huangqi-Gegen(Radix Astragali-Radix Puerariae)on indomethacin-induced gastric mucosa damage in rats. Methods SD rats were randomly divided into normal group,model group,positive drug group(cimetidine 12 mg·kg-1·d-1,ig),and high-,medium- and low-dose HuangqiGegen water decoction group(24, 12, 6 g·kg-1·d-1, ig) according to the body weight of rats. The intragastric administration for each group lasted 4 continuous days. Seventy-five minutes after the last medication,all rats except for the normal group were intraperitoneally injected with indomethacin(35 mg·kg-1)to induce gastric mucosa injury. Six hours after indomethacin treatment,the animals were executed for the sampling of the stomach. The mucosal injury was scored, and the pathological changes in gastric mucosa were observed. The contents of malondialdehyde(MDA),superoxide dismutase(SOD),and reactive oxygen species(ROS)in mucosal tissue were detected. Results The gastric mucosal injury of model group was severer than the normal group,showing as obvious gastric mucosal lesions and erosion,increased ROS and MDA contents,and decreased SOD activity. The drug pair groups at different dosages had an effect on relieving the mucosal injury and pathological changes,and decreased gastric mucosal injury scores as well as the MDA and ROS contents(P〈0.05,P〈0.01 compared with those in the model group). The high-dose drug pair enhanced SOD activity(P〈0.05 compared with that in the model group). Conclusion Drug pair Huangqi-Gegen exerts certain protective effect on indomethacin-induced gastric mucosa injury,and the mechanism may be related to the counteraction of oxidative stress.
